匹莫苯丹(pimobendan,PMBD)是国家二类新兽药。但由于该药的原料药成本较高,不易合成,反应步骤多而限制了该药物的临床普遍应用。文中提出了一种新的8步合成匹莫苯丹的新方法,以乙酰苯胺作为原料,通过8个步骤合成匹莫苯丹。化合物的每一步的产品纯度都可以达到95%以上,化合物的结构经核磁共振波谱(nuclear magnetic resonance hydrogen spectroscopy,NMR)和高分辨质谱(high resolution mass spectrometry,HRMS)确证。同时,新的方法条件温和,反应步骤少,操作简便,副反应少,纯度高,可以用于匹莫苯丹的放大合成。
Study on synthesis technology of pimobendan
Pimobendan(PMBD)is a new veterinary drug of national class Ⅱ.However,due to the high cost of the drug's raw material,difficult synthesis and many reaction steps,the clinical application of this drug is limited.In this paper,a new 8-step synthesis method of pimobendan was presented.Using acetanilide as raw material,pimobendan was synthesized in 8 steps.The purity of the product at each step of the compound can reach 95%or more.The structure of the compound was confirmed by nuclear magnetic resonance spectroscopy(NMR)and high resolution mass spectrometry(HRMS).At the same time,this new method has mild conditions,less reaction steps,simple operation,less side reaction and high purity.It can be used for amplifying synthesis of pimobendan.
