Effects of Berberine on Th1/Th2 Imbalance in Asthmatic Mice Based on Notch Pathway
Objective To explore the effect of berberine on Th1/Th2 imbalance in asthma model mice and its mecha-nism.Methods Asthma mouse model was induced by ovalbumin.Mice were divided into control group,model group,berberine group and berberine+inhibitor group.The inflammatory infiltration of lung tissue was observed by HE staining.The propor-tions of Th1(CD4+IFN-γ)and Th2(CD4+IL-4+)cells were detected by flow cytometry.The levels of IFN-γ and IL-4 in alveo-lar lavage fluid were detected by ELISA.The mRNA expressions of GATA3 and STAT6 in lung tissue were detected by qRT-PCR.The expression of Notch1 protein was detected by immunohistochemistry,and the expression levels of Notch1 and Hes1 proteins were detected by Western blotting.Results Compared with the control group,the aggregation of inflammatory cells in lung tissue of model mice increased,airway smooth muscle became thicker,the proportion of Th1 cells and IFN-γ level decreased significantly(P<0.05),while the proportion of Th2 cells,IL-4 level,GATA3 and STAT6 mRNA expression,Notch1 and Hes1 protein expression increased significantly(P<0.05).Compared with the model group,the aggregation of inflammatory cells in the lung tissue of berberine group decreased,airway smooth muscle became thinner,the proportion of Th1 cells and IFN-γ level increased significantly(P<0.05),while the proportion of Th2 cells,IL-4 level,GATA3 and STAT6 mRNA expression,Notch1 and Hes1 protein expression decreased significantly(P<0.05).Compared with berberine group,the effect of berberine was fur-ther strengthened in berberine+inhibitor group.Conclusion Berberine can restore Th1/Th2 balance and reduce the inflamma-tory injury of lung tissue in asthmatic mice,which may be related to activation inhibition of Notch signaling pathway.
