养血清脑醇提浸膏化学成分及抗氧化活性研究

目的:研究养血清脑醇提浸膏化学成分及其抗氧化活性.方法:采用D101大孔树脂柱色谱、硅胶柱色谱、十八烷基硅烷键合硅胶(ODS)柱色谱、制备型高效液相色谱(HPLC)等色谱分离技术进行分离纯化,运用质谱、核磁共振波谱等谱学技术鉴定化合物的结构,并通过清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基试验考察单体化合物的抗氧化活性.结果:从养血清脑醇提浸膏分离并鉴定19个化合物,分别鉴定为gluco-obtusin(1)、gluco-obtusifolin(2)、aurantio-obtusin(3)、senkyunolide I(4)、senkyunolide A(5)、neocnidilide(6)、obtusifolin(7)、chuanxiongnolide R1(8)、senkyunolide N(9)、senkyunolide J(10)、E-6,7-trans-dihydroxyligustilide(11)、chryso-obtusin-2-O-β-D-glucoside(12)、questin(13)、chrysoobtusin(14)、obtusin(15)、chrysophanol(16)、senkyunolide H(17)、Z-6-hydroxy-7-methoxy-glucilide(18)、3-butylidene-6α-hydroxy-7α-ethoxyphthalide(19).结论:首次对养血清脑醇提浸膏化学成分及抗氧化活性进行研究,分离得到的化合物均具有一定的抗氧化活性,其中senkyunolide I清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基的半数效应剂量(EC50)为89.71 μg/ml.
Study on the chemical composition and antioxidant activity of Yangxue Qingnao alcohol extract
Objective:To study the chemical components and antioxidant activity of Yangxue Qingnao alcohol extract.Methods:Chromatographic separation techniques such as D101 macroporous resin column chromatography,silica gel column chromatography,octadecylsilyl(ODS)column chromatography,and preparative high performance liquid chromatography(HPLC)for separation and purification.Spectral techniques such as mass spectrometry and nuclear magnetic resonance spectroscopy were used to identify the structure of the compound,and the antioxidant activity of the monomer compound was investigated through 1,1-diphenyl-2-picrylhydrazine(DPPH)radical scavenging test.Results:Nineteen compounds were isolated and identified from the alcohol extract of Yangxue Qingnao,which were identified as glucose obtusin(1),gluco-obtusifolin(2),aurantio-obtusin(3),senkyunolide I(4),senkyunolide A(5),sedanolide(6),obtusifolin(7),chuanxiongnolide R1(8),senkyunolide N(9),senkyunolide J(10),E-6,7-trans-dihydroxyligustilide(11),chryso-obtusin-2-O-β-D-glucoside(12),question(13),chrysoobtusin(14),obtusin(15),chrysophanol(16),senkyunolide H(17),Z-6-hydroxy-7-methoxy-glucilide(18),3-butylidene-6α-hydroxy-7α-ethoxyphthalide(19).Conclusions:The chemical composition and antioxidant activity of the brain alcohol extract from nourishing serum were studied for the first time.The isolated compounds all have certain antioxidant activity,among which senkyunolide I has good antioxidant activity.
