摘要

研究醴肠的化学成分和抗神经炎活性.本文采用D101大孔树脂、硅胶、Sephadex LH-20葡聚糖凝胶等柱色谱和半制备液相色谱等技术对鳢肠Eclipta prostrata 95%乙醇提取物的化学成分进行系统分离纯化,并结合核磁共振波谱数据以及与文献数据比对进行化合物的结构鉴定得到20个单体化合物,分别鉴定为:3,4-seco-olean-12-en-4-ol-3,28-dioic acid(1)、刺囊酸(2)、eclalbasaponin Ⅱ(3)、eclalbasaponin Ⅰ(4)、ecliptasaponin B(5)、eclalbasaponin Ⅳ(6)、eclalbasaponin Ⅵ(7)、eclalbasaponin Ⅶ(8)、香豌豆酚(9)、芹黄素(10)、蓟黄素(11)、川陈皮素(12)、木犀草素-7-O-β-D-葡萄糖苷(13)、金色鲜胺醇酯(14)、plantagoguanidinic acid(15)、plumbagine B(16)、蟛蜞菊内酯(17)、松脂醇(18)、α-醛基三聚噻吩(19)、α-terthienylmethanol(20).化合物1、14、15为首次从该植物中分离得到.之后利用脂多糖(lipopolysaccharide,LPS)诱导小鼠小胶质BV2细胞模型评价了部分化合物的抗神经炎活性,发现化合物15能显著减少LPS诱导BV2细胞中的NO释放量,IC50值为37.3 μmol/L,且无细胞毒性.胍类生物碱化合物15对LPS诱导的BV2细胞炎症具有一定的抗炎活性,且其抗炎活性为首次报道.

Abstract

This study aims to investigate the chemical constituents and inhibitory effect on neuroinflammation of Eclipta pros-trata.D101 macroporous resin,silica gel,Sephadex LH-20,and semi-prepared high-performance liquid chromatography were used to isolated and purified the chemical constituents of 95%ethanol extract Eclipta prostrata.Twenty monomer compounds were identified as 3,4-seco-olean-12-en-4-ol-3,28-dioic acid(1),echinocystic acid(2),eclalbasaponin Ⅱ(3),eclalbasa-ponin Ⅰ(4),ecliptasaponin B(5),eclalbasaponin Ⅳ(6),eclalbasaponin Ⅵ(7),eclalbasaponin Ⅶ(8),orobol(9),api-genin(10),cirsimaritin(11),nobiletin(12),luteolin-7-O-β-D-glucoside(13),aurantiamide acetate(14),plantagoguani-dinic acid(15),plumbagine B(16),wedelolactone(17),pinoresinol(18),α-formylterthienyl(19),and α-terthienylmeth-anol(20)based on the nuclear magnetic nuclear resonance(NMR)data and comparison with corresponding data of the re-ported literatures.Compounds 1,14,and 15 were isolated from E.prostrata for the first time.Some of the isolated compounds were assessed for their inhibitory effects on neuroinflammation by detecting NO level in BV2 cells induced by lipopolysaccha-ride(LPS),compound 15 significantly reduced NO release in LPS-induced BV2 cells with an IC50 of 37.3 μmol/L and showed no cytotoxicity.Guanidine alkaloid 15 was first reported with anti-inflammatory activity in LPS-induced BV2 cells.
