生物信息学评估肺腺癌中PD-L1表达对临床病理和预后的意义

目的 利用生物信息学评估肺腺癌中PD-L1表达的临床病理和预后意义,并且用免疫组化对其表达和临床病理进行验证.方法 利用GEPIA数据库分析PD-L1在数据库中mRNA的定量表达,并用免疫组化的方法进行验证;利用Te Kaplan-Meier Plotter评估PD-L1表达对肺腺癌预后价值的影响;利用Timer 2.0对肺腺癌的免疫细胞浸润进行分析;通过UALCAN数据库的TCGA模块,对不同亚型PD-L1在肺腺癌中的表达进行差异性分析.结果 生物信息学显示,PD-L1在癌组织中的表达明显低于正常组织,免疫组化结果显示,PD-L1在肺腺癌癌组织中呈高表达,但在正常组织中呈低表达或不表达.PD-L1的表达在不同年龄、性别、临床分期以及淋巴结转移中差异无统计学意义,但其表达与患者的总生存期、首次进展生存期显著相关.结论 PD-L1的表达与肺腺癌患者的生存预后有一定的关系.
Clinical pathological and prognostic significance of PD-L1 expression in lung adenocarcinoma in bioinformatics
Objective:To evaluate the clinical pathological and prognostic significance of PD-L1 expression in lung adenocarcinoma with bioinformatics,and to validate its expression and clinical pathology with immunohistochemistry.Methods:Using the GEPIA database to analyze the quantitative expression of PD-L1 mRNA in the database,and validate it with immunohistochemical methods;Using Te Kaplan Meier Plotter to evaluate the impact of PD-L1 expression on the prognostic value of lung adenocarcinoma;Analyzing immune cell infiltration in lung adenocarcinoma with Timer2.0;Differential analysis of the expression of different subtypes of PD-L1 in lung adenocarcinoma was performed by the TCGA module of the UALCAN database.Results:Bioinformatics showed that the expression of PD-L1 in cancer tissue was significantly lower than that in normal tissue.Immunohistochemical results showed that PD-L1 was highly expressed in lung adenocarcinoma tissue,but low or no expression was observed in adjacent tissues.The expression of PD-L1 was not significantly different by age,sex,clinical stage and lymph node metastasis,but its expression was significantly correlated with the overall survival OS and first progressive survival FP of patients.Conclusion:PD-L1 expression affects the survival prognosis of lung adenocarcinoma patients.
