单环刺螠体壁抗氧化肽酶解方法探究

摘要
目的 获得单环刺螠体壁抗氧化肽,探究最佳酶解工艺.方法 以单环刺蜢体壁为原料,以DP-PH自由基清除率为指标,在单因素实验基础上,利用响应面分析法,确定胰蛋白酶、木瓜蛋白酶和碱性蛋白酶的最佳水解条件.结果 筛选出三种酶制备抗氧化活性肽的最佳条件分别为:①胰蛋白酶:原料浓度0.8%,温度45℃,时间2.6 h,DPPH自由基清除率可达96.55%;②木瓜蛋白酶:原料浓度1.2%,温度55℃,时间3.3 h,DP-PH的自由基清除率可达99.99%;③碱性蛋白酶:原料浓度1.0%,温度54℃,时间2.8 h,DPPH自由基清除率可达97.40%.结论 以胰蛋白酶、木瓜蛋白酶和碱性蛋白酶的最佳水解条件制备抗氧化活性肽,其抗氧化率可达90%以上,为后续多肽的抗氧化机制、氨基酸序列研究以及单环刺螠肽的开发应用提供基础.
Abstract
Objective To obtain the antioxidant peptides from the body wall of Urechisunicinctus.Methods Using the body wall of Urechisunicinctus as the raw material and taking the DPPH radical scavenging ability as an in-dex,on the basis of single factor experiment and Response Surface Methodology.The product and the optimum prepara-tion conditions were obtained.Results Trypsin:substrate concentration 0.8%,temperature 45℃,time 2.6 h,DPPH radical scavenging ability can reach 96.55%.Papain:substrate concentration 1.2%,temperature 55 ℃,time 3.3 h,DP-PH radical scavenging rate can reach 99.99%.Alcalase:substrate concentration 1.0%,temperature 54℃,time 2.8 h,DPPH radical scavenging ability can reach 97.40%.Conclusion The antioxidant activity peptide was prepared under the optimum hydrolysis conditions of trypsin,papain and alkaline protease,and the antioxidant rate was over 90%.
