Bioinformatics analysis of key enzymes in chlorogenic acid anabolic pathway of Lonicera Japonica Thunb
4-coumaric acid coA ligase(4CL)is a key enzyme in the metabolic pathway of chlorogenic acid synthesis of honeysuckle. In this study, the physicochemical properties, systematic evolution and gene structure of the 4CL were analyzed.MEGA-11 software was used to compare the similarity and evolution-ary distance between amino acid sequences of the orthologs in plants and construct phylogenetic trees. The results showed that 4CL1 was composed of 538 amino acid residues, the molecular weight was 58.68 kDa,and the theoretical pI was 5.73.4CL2 is composed of 540 amino acid residues, the molecular weight can be measured to be 58.68 kDa,the theoretical pI value is 8.49.In addition, our results suggested that 4CL pro-tein is unstable protein, most of their amino acids are hydrophilic amino acids. This study can lay a theoreti-cal foundation for studying the biosynthesis mechanism of chlorogenic acid of honeysuckle and provide some reference for the application of chlorogenic acid metabolic engineering.
