Extraction and characterization of peach gum polysaccharide by high voltage pulsed electric field method
Peach gum is a gelatinous translucent substance rich in polysaccharides,which has a variety of biological activities,so it has high edible and medicinal value.In this study,peach gum polysaccharides were extracted from peach gum by high voltage pulsed electric field method.On the basis of single factor experiment,the optimal extrac-tion process was determined by orthogonal test as pulse voltage of 60 V,cycle number of 5 times,and solid-liquid ra-tio of 1∶100.Under these conditions,the content of the peach gum polysaccharide extracted was 94.56 g/100 g.Peach gum polysaccharides were characterized by molecular weight analysis,monosaccharide analysis,UV,FT-IR,XRD,SEM,Congo red experiment,CD and TGA.The results show that the gum polysaccharides are acidic polysac-charide with a pyranose ring composed of Ara,Gal,Xyl,Man and Glc-UA,and are amorphous polymer with tertiary helical structure.
