Effect of salidroside on angiogenesis and bone vessel coupling
Objective To study the effect of salidroside(SAL)on angiogenesis and bone vessel coupling.Methods MTS method was used to investigate the effect of osteoblast-conditioned medium on the proliferation activity of vascular endothelial cells.Tr-answell cell migration assay was used to investigate the effect of osteoblast-conditioned medium on the migration of vascular endothelial cells.Matrigel lumen formation experiment was used to investigate the effect of osteoblast-conditioned medium on lumen formation length.Fetal metatarsal angiogenesis assay was used to evaluate the effect of SAL on the sprouting area of endothelial cells.Results Compared with the conditioned culture medium group,the cell activity of the conditioned culture medium group after SAL treatment significantly increased,and the differences were statistically significant(P<0.01).The number of vascular endothelial cell migration and the length of HUVEC lumen formation in SAL group significantly increased compared with that in conditioned culture me-dium group,and the difference was statistically significant(P<0.01).The area of endothelial cell budding in SAl and vascular endo-thelial growth factor(VEGF)treatment groups was significantly larger than that in the control group,with statistical significance(P<0.01),and the above effects could be blocked by the anti-VEGF antibody Avastin.Conclusions SAL is a good angiogenic drug mon-omer,and can promote bone vascular coupling by up-regulating the expression of VEGF.
