摘要

目的 探讨氯化锰(MnCl2)对于原代星形胶质细胞的影响.方法 原代分离星形胶质细胞,培养后用星形胶质细胞特异性蛋白胶质纤维酸性蛋白(GFAP)的抗体鉴定细胞,用不同剂量的MnCl2处理24 h后MTT法检测星形胶质细胞的存活率,高内涵分析仪检测细胞面积和GFAP的表达,流式细胞仪检测细胞内ROS的变化.结果 原代分离的细胞鉴定为星形胶质细胞,不同剂量的MnCl2处理24 h后,星形胶质细胞的存活率随MnCl2剂量的升高呈现剂量依赖性下降,细胞面积缩小,GFAP表达增加,细胞内ROS增多.结论 MnCl2可以导致星形胶质细胞的过度活化.

Abstract

Objective To explore the influence of manganese to astrocyte cells.Methods Primary astrocytes were dissociated from newborn rat,and identified with antibody of glial fibrillary acidic protein (GFAP),which is specific protein expressed in astrocytes.Astrocytes were treated with different concentrations of manganese chloride (MnCl2) for 24 h.The survival rate of astrocytes were detected with MTF colorimetry test.The cell areas and expression of GFAP were measured by high content analyzer,and the ROS was determinated with flow cytometry.Results Mter 24 h treated with different concentrations of MnCl2,the survival rate of astrocytes decreased dosedependently,the areas of astrocytes were reduced,the expression of GFAP and the ROS increased.Conclusion Manganese chloride induces the activation of astrocytes.
