摘要

目的 开展邻苯二甲酸二环己酯(dicyclohexyl phthalate,DCHP)细胞毒性的研究,为科学评估DCHP的人群健康风险提供理论依据和数据支持.方法 基于高内涵细胞分析系统,利用人乳腺管癌细胞T47D和大鼠心肌细胞H9c2,开展DCHP对这2种细胞细胞活性、细胞凋亡和细胞核变化的检测.结果 DCHP明显提升T47D细胞的细胞活性(24和48 h F值分别为68.038和17.875,P<0.05),增加细胞核的面积,未导致较高的细胞凋亡率;DCHP明显降低H9c2的细胞活性(24和48 h F值分别为96.194和16.857,P<0.05),减少了细胞核面积,同时也导致了较高的细胞凋亡率.结论 DCHP具有较明确的雌激素活性,同时对心血管疾病的风险也需进一步关注.

Abstract

Objective To carry out the study on the cytotoxicity of dicyclohexyl phthalate(DCHP)and provide theoretical basis and data support for scientific assessment of the population health risk of DCHP.Methods DCHP was used to detect the cell viability,apoptosis,and nuclear area changes to human breast cancer cell T47D and rat myocardial cell H9c2 with a high connotation cell analysis system.Result DCHP significantly increased the cell activity of T47D cells and increased the nuclear area of T47D cells(The F-values for 24 and 48 hours are 68.038 and 17.875,P<0.05,respectively),but the cell apoptosis rate was low;DCHP significantly reduced the cell activity of H9c2(The F-values for 24 and 48 hours are 96.194 and 16.857,P<0.05,respectively)and significantly reduced the nuclear area,leading to a higher rate of cell apoptosis.Conclusion DCHP has clear estrogen activity,and further attention is needed to address the risk of cardiovascular disease.
