The cytotoxicity in vitro and the intradermal reaction test in rabbits of a novel denture material
Objective A preliminary safety assessment of a novel denture material was conducted through in vitro cytotoxicity and intradermal reaction tests.Methods After maceration in 10%serum medium,sterilized saline,and sterilized cottonseed oil,respectively,the JSAM-BL material and two types of 3D-printed samples underwent in vitro cytotoxicity test and intradermal reaction test according to national standards.Results In the cytotoxicity test in vitro,JSAM-BL material,JSAM-SLASH2-D crown,and JSAM-TL3-D crown were macerated in 10%serum medium.The cell viability rates of the three samples in 100%maceration fluid were 2.40%,28.08%,and 73.92%,respectively.In the intradermal reaction test on rabbits,the polar test groups for JSAM-BL material,JSAM-SLASH2-D crown,and JSAM-TL3-D crown received final scores of 0.20,0,and 0,respectively,while the non-polar test groups received final scores of 0,0,and 0.04,respectively.Conclusion The result of the in vitro cytotoxicity test indicated potential cytotoxicity for JSAM-BL material and JSAM-SLASH2-D crown,while JSAM-TL3-D crown showed no cytotoxicity.The intradermal reaction test on rabbits demonstrated no dermal reactions for both polar and non-polar maceration fluids of JSAM-BL material,JSAM-SLASH2-D crown,and JSAM-TL3-D crown.
