The effect of mongolian drug Huricha-6,Gurigumu-13 on allergic rhinitis model mice
Objective To observe the effects of Mongolian Huricha-6 and Gurigumu-13 on nasal mucosal lesions and IgE,IL-4 and IFN-γ content in allergic rhinitis(AR)mice.Methods 50 healthy BALB/c mice were divided into blank control group,AR model group,cetirizine group,Huricha-6 group and Gurigumu-13 group.AR mouse model was established,the behavioral score of each experimental group was scored,and one blank group and one mouse were randomly selected for passive skin allergen experiment(PCA).After treatment,administration,the pathological histological changes in the nasal mucosa were observed and the amounts of IgE,IL-4 and IFN-γ in serum were determined.Results(1)The successful AR model mice developed symptoms such as severe nose scratching and sneezing,and scored 5 points;after treatment,the nasal symptoms decreased and the symptom score decreased.(2)The PCA of the mice in the selected model group was positive,and no blue formation was found in the blank group,assuring the successful modeling of the mice in the model group.(3)Histological observation of the nasal mucosa in the AR model group showed more inflammatory cell infiltration,more interstitial widening,and dilated and more small blood vessels.After treatment,the symptoms of mice in each treatment group were relieved and inflammatory cells were decreased in the nasal mucosa,among which the Huricha-6 group was the most significant.(4)The serum contents of IgE and IL-4 in the model group were significantly higher compared with the blank control group,and the content of IFN--γ was relatively low(P<0.05).After treatment,the nasal symptoms and the content of IgE and IL-4 decreased significantly compared with the model group,and the content of IFN-γ increased(P<0.05).Between each group and cetirizine,there was no significant difference in IgE(F=0.098,P>0.05),IL-4(F=0.316,P>0.05),IFN-γ was statistically different(F=5.177,P<0.05),and no gender difference in each group(P>0.05).Conclusion Mongolian Huricha-6 and Gurigumu-13 can effectively improve the typical symptoms such as nose scratching,sneezing,runny nose and mental distress in allergic rhinitis model mice,promote nasal mucosa repair and reduce the pathological damage of AR mice;by regulating immune-related cytokines IgE,IL-4 and IFN-γ,it can effectively treat allergic rhinitis,lay a theoretical basis for exploring the related mechanism of Mongolian therapy in AR,and provide a research basis for future screening of AR targets and targeted therapy.
