目的 通过建立青春期雄性小鼠电子烟烟雾暴露模型,研究电子烟烟油主要成分对青春期雄性小鼠睾丸组织结构与功能的影响.方法 100只28 d龄SPF级雄性KM小鼠随机分为空白对照组、电子烟低剂量组、中剂量组、高剂量组及香烟组,采用呼吸道静式染毒,每日2次,每次30 min,连续染毒14以及28 d,检测睾丸及附睾脏器系数、精子数量、活动度及畸形率,测定血清激素水平以及睾丸组织氧化应激水平,苏木素-伊红(HE)染色观察睾丸组织学变化,TUNEL染色观察睾丸组织细胞凋亡情况.结果 与对照组比较,暴露于电子烟烟雾中的小鼠睾丸脏器系数上升(F14 d=1.240,F28 d=5.382,P<0.05),附睾脏器系数下降(F14 d=1.675,F28 d=5.152,P<0.05);附睾精子异常率增加(F精子数量(14 d)=6.583,F精子活动度(14 d)=54.996,F精子畸形率(14 d)=258.404,F精子数量(28 d)=3.872,F精子活动度(28 d)=63.392,F精子畸形率(28 d)=554.258,P<0.05);血清中睾酮(testosterone,T)、促黄体生成激素(luteotropichormone,LH)、促卵泡生成激素(follicle stimulating hormone,FSH)水平均降低(FT(14 d)=79.903,FLH(14 d)=88.929,FFSH(14 d)=258.201,FT(28 d)=90.364,FLH(28 d)=286.284,FFSH(28 d)=195.418,P<0.05);睾丸组织中 SOD 活力在 14 d 时上升(F14 d=3.275,P<0.05),28 d 时下降(F28 d=59.602,P<0.05),GSH-Px 活力下降(F14 d=2.645,F28 d=2.406,P>0.05),MDA 含量上升(F14 d=16.807,F28 d=53.520,P<0.05);病理检测无明显组织学变化;细胞凋亡阳性率增加(F14 d=927.885,F28 d=347.032,P<0.05).结论 电子烟烟油主要成分会导致小鼠睾丸肿胀,附睾萎缩,精子数量减少、活动度下降、畸形率上升;这可能与电子烟烟油主要成分致血清激素水平降低,睾丸组织发生明显脂质过氧化及生殖细胞凋亡有关.
Effect of the main components of e-cigarette e-liquid on the structure and function of testicular tissue in adolescent male mice
Objective The effects of e-cigarette e-liquid components on testicular structure and function of adolescent male mice were studied by establishing e-cigarette smoke exposure model.Methods One hundred 28-day-old SPF-grade male KM mice were randomly divided into blank control group,e-cigarette low-dose group,medium-dose group,high-dose group and cigarette group.The mice were exposed to respiratory static poison twice a day for 30 minutes each time for 14 days and 28 days,and the coefficient of testis and epididymis organs,sperm count,mobility and malformed rate were detected.The levels of serum hormone and oxidative stress in testicle tissue were measured.The histological changes were observed by HE staining.The apoptosis of testicular tissue was observed by TUNEL staining.Results Compared with the control group,the testicular organ coefficient of mice exposed to e-cigarette smoke increased(F14 d=1.240,F28 d=5.382,P<0.05),and the epididymal organ coefficient decreased(F14 d=1.675,F28 d=5.152,P<0.05).The abnormal sperm rate of epididymis increased(FSperm count(14d)=6.583,F Sperm motility(14d)=54.996,FSperm malformation rate(14d)=258.404,FSperm count(28 d)=3.872,FSperm motility(28d)=63.392,FSperm malformation rate(28 d)=554.258,P<0.05).Serum levels of testosterone,luteinizing hormone and follicle stimulating hormone were decreased(FT(14 d)=79.903,FLH(14 d)=88.929,FFSH(14 d)=258.201,FT(28 d)=90.364,FLH(28 d)=286.284,FFSH(28 d)=195.418,P<0.05).SOD activity increased at day 14(F14 d=3.275,P<0.05),and decreased at day 28(F28 d=59.602,P<0.05),GSH-Px activity decreased(F14 d=2.645,F28 d=2.406,P>0.05),MDA content increased(F14 d=16.807,F28 d=53.520,P<0.05).There were no obvious histological changes in pathological testing.The positive rate of apoptosis was increased(F14 d=927.885,F28 d=347.032,P<0.05).Conclusion The main components of e-cigarette e-liquid can cause testicular swelling,epididymis atrophy,reduced sperm count,decreased mobility,and increased deformity rate in mice.This may be related to the decrease of serum hormone level caused by the main components of e-cigarette e-liquid,the significant lipid peroxidation of testicular tissue and germ cell apoptosis.
