Effect of Wen Radix Codonopsis polysaccharide on intestinal mucosal immune function in mice
Objective To study the effect of Wen Radix Codonopsis polysaccharide on intestinal mucosal immune function in immunosuppressed mice.Methods Sixty Balb/c female mice were randomized into six groups:positive control group,blank group,model group,low dose polysaccharide group,medium dose polysaccharide group and high dose polysaccharide group,with 10 mice in each group.Except the blank group,other groups were injected intraperitoneally with cyclophosphamide to replicate the immunosuppressive model.After successful modeling,the positive control group was given Sijunzi decoction,model group and blank group were given distilled water,the low-,medium-and high-dose group were given corresponding concentration of codonopsis polysaccharide,each group was given continuously gavage for 14 days.The changes of body weight,thymus index and spleen index were measured,and the number of PPs in the whole small intestine were counted;The positive expressions of IgA,IL-2,IFN-γ and CD3+,CD4+,CD8+T cell subsets on intestinal mucosa were detected and analyzed by immunohistochemistry.Results Compared with the blank control group,the spleen index,thymus index,PPs number,CD3+,CD4+T lymphocyte synthesis,IgA,IL-2,IFN-γ secretion in the mouse in the model group were significantly reduced(P<0.05),and the immunosuppression mouse model was successfully established.Compared with the model group,the positive control group and the medium dose spleen index(F=8.574,P<0.05),the thymus index(F=9.705,P<0.05),the number of PPS(F=13.227,P<0.05),IgA(F=9.029,P<0.05),IL-2(F=6.492,P<0.05),IFN-γ(F=13.469,P<0.05),CD3+(F=6.164,P<0.05),CD4+(F=7.741,P<0.05),CD8+(F=5.787,P<0.05)has increased significantly,while the high dose inhibited the increase in mouse thymus index,the spleen index,and the synthesis of CD3+T,CD8+T,IgA and IFN-γ.Conclusion Within a certain dose range,Wen Radix Codonopsis polysaccharide can significantly enhance the intestinal mucosal immune function of mice,the medium dose was the best.In contrast,high dose reduced or inhibited intestinal mucosal immune function in mice.
