Optimization of Crude Polysaccharide Extraction Process and Antioxidant Activity of Bobai Zhi Using Response Surface Ultrasound Assisted Method
In order to obtain the optimal extraction process and antioxidant parameters of crude polysaccharides from Bobai Zhi.Using ultrasound assisted design,a three factor and three level response surface experiment was conducted to optimize the extraction process of polysaccharides from Bobai Zhi;And the scavenging ability of DPPH and·OH free radicals was measured to study the antioxidant activity of Bozhou Baizhi crude polysaccharide.The results showed that the highest polysaccharide extraction rate was 8.65%at a liquid to material ratio of 59:1(mL/g),ultrasound temperature of 72℃,ultrasound time of 81 min,and ultrasound power of 250W;The concentration of Bai Zhi polysaccharide shows a dose-response relationship with free radical scavenging rate within a certain range.When the polysaccharide concentration is 4mg/mL,the hydroxyl radical scavenging rate is 94.39%,and the DPPH free radical scavenging rate can also reach 85.07%,which is close to the VC positive control group.Research has shown that the response surface ultrasound assisted optimization of the extraction process for crude polysaccharides from Angelica dahurica is feasible.Angelica dahurica polysaccharides have good antioxidant activity and can be used as natural antioxidants.
