Reducing Carbon Source Combination of Bacillus licheniformis ACCC 01957
Bacillus licheniformis is commonly used in fermented feed.This experiment aims to improve the biomass and spore yield of B.licheniformis strain ACCC 01957 by optimizing the most important factor,carbon source,in the fermentation process.The utilization efficiency of 71 carbon sources on Biolog GenⅢ plate was firstly screened,the effect of favorable carbon sources on the biomass,growth cycle and sporulation rate was then analyzed by using an au-tomatic cell growth analyzer,and the optimal fermentation conditions were determined on the flask level.The results showed that ACCC 01957 made better use of 25 carbon sources,and N-acetyl-β-D-mannosamine,D-galactose,L-rhamnose,inosine,glycerin,pectin,L-lactic acid,Tween 40,L-aspartic acid,L-glutamic acid and L-pyroglutamic acid simultaneously promoted the growth and sporulation;sorbitol was identified as the carbon source for promoting growth and inhibiting spore production;when glucose and L-rhamnose were combined at the ratio of 1∶6 in shaking flask level,the biomass at 24 h was the highest(8.3 × 10 8 cfu/mL),and the sporulation rate at 48 h reached the maximum(91.19%).This study greatly improved the growth,spore yielding and sporulation rate of B.licheniformis ACCC 01957 by optimizing the carbon source.
