The vast majority of the Earth's microbes remain uncultured.Isolation and cultivation of microorganism are beneficial to the discovery and application of new species resources and new natural products,as well as the develop-ment of basic researches on the physiology and biochemistry,metabolic potential,evolution and ecological function of microorganism.Activation and enrichment are imperative for the isolation and cultivation of microorganism in natural habitats.In this study,the effects of different media(WB,PJ,WB/10 and PJ-M),different activation durations(1-29 d),and different activation conditions(aerobic and anaerobic)on microbial activation and enrichment of soil sam-ples were analyzed by 16S rRNA gene V3-V4 amplicon sequencing and metagenomic sequencing.It was found that the different activation media,cultivation conditions and activation time significantly affected the composition of microbial population.Most of which the use of medium with relatively low nutrient concentration(WB/10)and extension of ac-tivation time(5-29 days of activation)conducive to improve the diversity of activated microorganism,the activation,and the detection as well as CPR and other of microbial dark matter.It has certain guiding effect on the subsequent isolationand cultivation of microorganism,especially microbial dark matter.
