以种子和叶片为外植体,研究了离体条件下外植体类型及植物生长调节剂对山桐子器官发生和植株再生的影响.结果表明:种子为诱导愈伤组织的适宜外植体;在MS+1.0 mg·L-16-BA +1.0mg· L-1 NAA +3%蔗糖+0.6%琼脂的培养基上,愈伤诱导率达96.8%;继代培养在MS+0.08 mg·L-1 TDZ+1.5 mg·L-16-BA +0.05 mg·L-1 NAA培养基上能成功的诱导芽的分化,诱导率达48.3%以上;芽增殖的适宜培养基为1/2MS +0.03 mg·L-1 TDZ +2.0 mg·L-16-BA +0.1 mg·L-1 NAA +3%蔗糖+0.6%琼脂,增殖系数为4.83;在1/2MS+0.5 mg·L-1 IBA +2%蔗糖+0.6%琼脂培养基上,无根苗生根率达88%.
Establishment of Plantlet Regeneration System of Idesia polycarpa in vitro
Seeds and tender leaves of Idesia polycarpa were used as a source of initial explants.The effects of explants and plant growth regulators on organogenesis and plantlet regeneratio in vitro were studied.The results showed that seeds were more suitable for initial explants in inducing calli.High induction rate of 96.8% was achieved in MS medium supplemented with 1.0 mg· L-1 NAA,1.0 mg· L-1 6-BA,3% sucrose and 0.6 %agar.MS medium supplemented with 0.08 mg · L-1 TDZ,1.5 mg · L-1 6-BA and 0.05 mg · L-1 NAA in subculture was in favor of the shoot differentiation and the rate was over 48.3%.The shoot proliferation coefficient reached 4.83 in 1/2MS medium with 0.03 mg · L-1TDZ,2.0 mg · L-1 6-BA,0.1 mg · L-1 NAA,3% sucrose and 6% agar.High rooting percentages and excellent development root systems were obtained on 1/2MS medium with 0.5 mg · L-1 IBA,2% sucrose and 0.6% agar.
