Folin-Ciocalteu法测定大熊猫主食竹中多酚含量

为建立Folin-Ciocalteu法测定大熊猫主食竹缺苞箭竹(Fargesia denudata)总酚酸含量的方法.以没食子酸为对照品,研究了Folin-Ciocalteu法测定缺苞箭竹中酚酸类物质含量的适宜条件.结果表明,适宜的比色条件为Folin-Ciocalteu试剂与10% Na2CO3体积比为1∶6,Folin-Ciocalteu试剂3 mL,显色温度25℃,反应时间2h,检测波长780 nm.浓度为20~100 μg·mL-1时与其吸光值呈良好的线性关系,测定方法的平均回收率为99.4%.可见该法有较好的准确度和重现性.
Determination of the Contents of Polyphenols in Fargesia denudata, the Main Food of Giant Penda by Folin-Ciocalteu Colorimetric Method
Aims:to establish a quantitative method of measuring total phenols in Fargesia denudata,the main food of giant panda.Methods:with gallic acid as reference substance to study the suitable conditions of the measurement by Folin-Ciocalteu method.Results:the optimum conditions for determining total polyphenols were as follows:the volume ratio of Folin-Ciocalteu reagent to 10% Na2CO3:1 ∶ 6,3 mL of Folin-Ciocalteu reagent incubated at 25℃for 2 h,followed by detection at 780 nm.Good linear relationship was found in the concentration of 20-100μg · mL-1,the average recovery was 99.4%.
