骏枣组培快繁体系及菌根化育苗技术研究

骏枣是我国主栽制干枣品种,但基于组织培养技术的高效快繁育苗技术体系尚未建立。本文围绕骏枣的腋芽增殖、继代、生根培养基筛选以及生根组培苗的菌根化开展研究。结果表明:在改良MS上添加1。0 mg/L 6-BA、0。2 mg/L IBA以及0。012 mg/L TDZ可有效提高骏枣茎段不定芽增殖效率,增殖系数达4。26。在改良MS培养基中添加1。2~1。5 mg/L 6-BA+0。2 mg/L IBA为继代培养基时,增殖和生长状态最佳。在生根培养阶段,茎段在1/2MS培养基中添加0。6 mg/L IBA和0。5%的活性炭,不定芽的生根率达到86。89%。在生根组培苗移栽时接种含有异形根孢囊霉孢子的内生菌根菌剂,可显著提高其移栽成活率和生物量,接种菌剂的生根苗成活率达96。30%,较对照高出18。52%,菌根接种组平均苗高为8。1 cm,为对照的1。59倍。本研究所建立的骏枣组培快繁体系和菌根化育苗技术,增殖效率高,移栽后成活率高、生长速度快,可获得大量生长健壮的植株,为实现骏枣种苗规模化繁殖提供完善的技术体系,为骏枣的规模化育苗提供技术支持。
Establishment of Rapid Propagation in vitro and Mycorrhizal Seedling System of Ziziphus jujuba'Junzao'
'Junzao'jujube(Ziziphus jujuba'Junzao')is a dominant dry jujube cultivar in China,the effi-cient rapid propagation technology has not yet been established well.In this paper,focused on the related issues of axillary bud proliferation,subculture,selection of rooting medium,and mycorrhization of rooted seedlings from tissue culture,we tried to establish an efficient and stable propagation technique.The results showed that adventitious buds could be propagated effectively on a modified MS supplemented with 1.2 mg/L 6-BA+0.2 mg/L IBA+0.012 mg/L TDZ.The shoot could be sub-cultured on modified MS supplemented with 1.2-1.5 mg/L 6-BA+0.2 mg/L IBA.Root could be induced efficiently on the optimal rooting medium(1/2MS+IBA 0.6 mg/L+sucrose15 g/L),the root induction rate of shoots was up to 86.89%.Transplanting rooting seedlings into pots containing arbuscular mycorrhizal agents promoted seedling growth significantly compared with non-inoculated seedlings,the survival rate reached 96.30%,18.52%higher than that of the control.The average height of inoculated seedlings was 8.1 cm,1.59 times higher than that of the control.In conclusion,the rapid propagation system in vitro and mycorrhizal seed-ling technology have been established manifested by the features of high proliferation rate,high survival rate after transplantation,and high seedling growth rate,from which a large number of healthy jujube seed-lings can be produced.The results provide technical support for large-scale jujube seedling cultivation.
