Isolation and Identification of Pathogens Causing Stem Rot of Cistanche deserticola in Qingtu Lake,Minqin
To clarify pathogen species causing stem rot in Cistanche deserticola at Qingtu Lake of Minqin,Gansu Province,the tissue isolation method,morphological characteristics and ITS molecular methods were used for pathogen isolation and identification.The results showed that three different pathogens were isolated from stem rot in C.deserticola at Qingtu Lake,the symptoms observed both in indoor and field were essentially the same,showing typical characteristics of stem rot.These patho-gens were designated as RCRJF-1,RCRJF-2,and RCRJF-3,respectively.The ITS molecular identifica-tion showed that RCRJF-1,RCRJF-2 and RCRJF-3 exhibited 100%similarity to Fusarium prolifera-tum(accession number MT560212),Fusarium oxysporum(accession number MT530243)and Fusar-ium acuminatum(accession number MH341217),respectively.Based on morphological characteris-tics,it was determined that the pathogens causing the stem rot in C.deserticola in Qingtu Lake,Min-qin,Gansu were F.proliferatum,F.oxysporum,and F.acuminatum.The isolation frequencies were 52.38%,31.11%,and 16.51%,respectively,with F.proliferatum identified as the dominant pathogen.
