Establish the method for determining the content of triolein in semen coicis.Ultrasonic strengthening extraction was adopted,high performance liquid chromatography was used for determination.Chromatographic conditions:Separations were performed on a Diamonsil C18 column (250 mm×4.6 mm,5μm) with an isocratic mobile phase of acetonitriledichloromethane (60 ∶ 40),the flow rate was 1.0 mL/min and the evaporative light scattering detector was used,the tube temperature was 70 ℃ and the pressure of nitrogen was 35 psi.The calibration curve was liner over the range of 0.241～2.410μg for triolein (r=0.9963),the average recovery was 101.42％ and the relative standard deviation (RSD) was 2.6％(n=3).The method is simple accurate with a good reproducibility and can be applied to quality of semen coicis.
