作为一种新的细胞程序性死亡方式,细胞焦亡(pyroptosis)在许多疾病中起着重要作用.近年来,细胞焦亡与肥胖及其代谢综合征之间的关系越来越受到关注.大量研究表明NLRP3炎性体介导的细胞焦亡可影响肥胖和代谢相关综合征的进展,而靶向NLRP3炎性体则具有潜在的治疗效果.该文首先全面阐述了NLRP3炎性体介导细胞焦亡的相关分子机制;其次,基于现有证据,讨论了NLRP3炎性体介导的肥胖和代谢综合征的最新进展;最后,还列出了一些具有靶向抑制NLRP3炎性体激活的药物和制剂,这可能为预防治疗肥胖和相关代谢综合征及药物开发带来更多选择和益处.
Research Progress of NLRP3 Inflammasome in Obesity and Its Related Metabolic Syndromes
As a new form of cell programmed cell death,pyroptosis plays an important role in many dis-eases.In recent years,the relationship between pyroptosis and obesity and its related metabolic syndromes receives much more attention.A large number of studies have shown that NLRP3 inflammasome-mediated pyroptosis can affect the progression of obesity and its related metabolic syndromes.Therefore,the NLRP3 inflammasome is a promising target with great therapeutic potential for treating obesity and its associated diseases.In this article,firstly the molecular mechanism of NLRP3 inflammasome mediated pyroptosis was described.Secondly,this article discussed the latest advances in NLRP3 inflammasome in obesity and its related metabolic syndromes.Finally,a number of drugs and formulations with targeted inhibition of NLRP3 inflammasome activation are listed,which may lead to more options and benefits for preventing and/or treating obesity and related metabolic syndromes.
