Significance of methylation of SHOX2 and RASSF1A genes in patients with benign lung disease
Objective To analyze the methylation status of SHOX2 and RASSF1A in paraffin samples of patients with benign lung diseases and explore their clinical significance.Methods This study selected 44 patients diagnosed with benign lung diseases from West China Hospital of Sichuan University from 2018 to 2019 as archive samples,including 31 paraffin tissue samples and 13 cell block samples.Real time fluorescence PCR was used to detect methylation of SHOX2 and RASSF1A genes,and positive results were followed up by telephone for a period of 3 years,with the endpoint being clinically confirmed lung cancer.Results The positive rate of SHOX2 methylation in the selected samples(29.27%)was significantly higher than that of RASSF1A methylation(4.88%),and the difference was statistically significant(P<0.05).The positive rate of SHOX2 methylation in organizational samples(32.26%)was significantly higher than that of RASSF1A methylation(3.23%),and the difference was statistically significant(P<0.05).Comparing the methylation positivity rate between tissue samples and cell block samples,it was found that the former had a slightly higher methylation positivity rate(35.48%)than the latter(20.00%).There was no simultaneous positive result for both genes in the organizational sample,while there was one case in the cell block sample where both genes were simulta-neously positive.There was no statistically significant difference between the methylation results of SHOX2 and RASSF1A and the pathological type(P>0.05).Telephone follow-up was conducted on 11 methylation positive patients,of which 1 patient was clinically diagnosed with lung cancer.Conclusion SHOX2 and RASSF1A methylation positive cases exist in patients with benign lung diseases,and follow-up revealed that some patients have developed lung cancer.Therefore,when the DNA methylation test result is positive in patients with benign lung diseases,combined with clinical characteristics,follow-up is necessary,which is conducive to early detection and treatment of possible tumor progression.
