Relationship between the expression of PTEN and 6KDMB and clinical pathological characteristics in lung adenocarcinoma
Objective To observe the expression of PTEN and 6KDMB in lung invasion adenocarcinoma,analyze the correlation between the expression of PTEN and 6KDMB with clinicopathological parameters and their relationship,and deepen our understanding of the pathogenesis of lung adenocarcinoma.Methods The expression of PTEN and 6KDMB mRNA in lung adenocarcinoma and normal lung tissues was evaluated by bioinformatics analysis.Immunohisto-chemical SP method was used to detect the expression of PTEN and 6KDMB protein in 74 cases of lung invasive adeno-carcinoma and corresponding normal lung tissues,and the relationship between the two protein and clinicopathological pa-rameters of lung invasive adenocarcinoma was analyzed.The correlation between the two was analyzed by Spearman method.Results The expression levels of PTEN and 6KDMB mRNA in invasive lung adenocarcinoma tissues were down-regulated compared with normal lung tissues.The results of analysis of 59 paired lung adenocarcinoma and normal lung tissues showed that the expression level of 6KDMB mRNA in lung adenocarcinoma tissues was down-regulated com-pared with that in normal lung tissues(P<0.05).Immunohistochemical results showed that the expression rate of PTEN protein in invasive lung adenocarcinoma(33.78%,25/74)was significantly lower than that in normal lung tissue(54.05%,40/74),and the expression rate of 6KDMB protein(72.97%,54/74)was significantly higher than that in normal lung tissue(21.62%,16/74)(P<0.05).Among 74 cases of invasive lung adenocarcinoma,PTEN had a higher expression rate(P<0.05)with high differentiation,no airway dissemination,and TNM stage Ⅰ+Ⅱ expression.6KDMB had a higher expression rate in lymph node metastasis and TNM stage Ⅲ+Ⅳ(P<0.05).The lower the expres-sion of PTEN and the higher the expression of 6KDMB in 74 cases of invasive lung adenocarcinoma,and the Spearman correlation analysis showed a significant negative correlation between PTEN and 6KDMB(Rs=-0.53,P<0.001).Conclusion PTEN and 6KDMB proteins in invasive lung adenocarcinoma are correlated with invasion and metastasis,and they are significantly negatively correlated.The study on the expression and correlation of PTEN and 6KDMB in lung in-vasive adenocarcinoma will provide a theoretical basis for further study of molecular biological mechanisms.
