Effect of Fangfeng Tongsheng Granule on gut microbiota and related inflammatory factors in mice with chronic urticaria
Objective To explore the effect of Fangfeng Tongsheng Granule on allergic reaction in chronic urticaria(CU)mice model and its possible mechanism.Methods 60 male BALB/c mice of SPF grade were selected and divided into 6 groups according to random number table:blank control group,model group,levocetirizine group,high,medium and low dose groups of Fangfeng Tongsheng Granule,with 10 mice in each group.The CU mouse model was constructed and treated for 21 days.The general characteristics of the mice were observed and the allergic reaction intensity of the mice was scored.Determination of IL-17 and IFN-γ in mouse serum by enzyme-linked immunosorbent assay-y Level.The changes of intestinal flora were observed by 16S rRNA sequencing.Results Compared with the blank control group,the allergic reaction score of the model group mice increased significantly(P<0.01).IL-17 and IFN in serum of CU mice were increased,including IFN-γ significantly higher(P<0.01).16S rRNA sequencing analysis showed that there was no significant change in the abundance index and diversity index of intestinal flora after intervention.The spe-cies composition and LEfSe analysis showed that compared with the model group,the high dose group of Fangfeng Tong-sheng Granule had an increase in the relative abundance of Achromobacter species.The relative abundance of Proteobac-teria,Campylobacter and Spirobacteriaceae decreased.PICRUSt2 function prediction analysis showed that the intestinal flora of mice in each group was mainly related to carbohydrate metabolism,amino acid metabolism and other metabolic pathways.Conclusion Fangfeng Tongsheng Granule can significantly reduce the allergic reaction of CU mice,and its mechanism may be related to the reduction of IL-17 and IFN-γ.It is related to the expression and regulation of the com-position and structure of intestinal flora and the improvement of its metabolic pathway.
