Chemical constituents of Lonicera caerulea var.edulis and their cytotoxicity in HepG2 cells
Objective To study the chemical constituents of Lonicera caerulea var.edulis and their cytotoxic activities in HepG2 cells.Methods The extract of L.caerulea was isolated and purified by normal phase column chromatography,reverse phase column chro-matography,Sephadex LH-20 chromatography,and HPLC,etc.The structures of compounds were elucidated by detailed spectro-scopic analysis based on physicochemical properties of the compounds.MTT method was used to evaluate their cytotoxicity in HepG2 cells.Results Twelve compounds were isolated and determined as ferna-7,9(11)-diene-3α,16α-diol(1),3β,23-dihydroxy-olean-12-en-28-oic acid(2),2-(3-methoxy-4-hydroxyphenyl)-propane-1,3-diol(3),nicotinic acid(4),4-hydroxy-5-methoxy-benzoic acid(5),3,7-dihydro-1,3,7-trimethyl-1H-purine-2,6-dione(6),caffeic acid methyl ester(7),(2E)-2,6-dimethyl-6-hy-droxy-2,7-octadienoic acid(8),syringin(9),threo-5-hydroxy-3,7-dimethoxyphenylpropane-8,9-diol(10),3,4,5-trimethoxyphe-nyl 1-α-glucofuranoside(11),and benzyl-β-D-glucoside(12).Among these compounds,compound 1 exhibited some cytotoxicity in HepG2 cells.Conclusion The identified compounds mainly contained 2 tritepenoids,2 alkaloids,3 lignins,one unsaturated fatty acid,and 4 aromatic compounds.Compounds 3,8,10,11 were isolated from Lonicera genus for the first time,and 1,2,4,6,7 and 12 were isolated from the species for the first time.
