Effect of Poria cocos polysaccharides on autoimmune inflammatory myopathy in rats
Objective To investigate the effect of Poria cocos polysaccharide(PCP)on the immune system of autoimmune inflammatory myopathy(IIM)of rats and its possible mechanism.Methods A total of 40 IIM model rats were randomly divided into model group,PCP low-dose,PCP medium-dose and PCP high-dose groups,10 rats in each group,and also a control group(10 rats)was set up.Rats in low,medium and high dosage of PCP groups were administered by gavage with 75,150 and 300 mg·kg-1 PCP solution(prepared with distilled water),and the control group and model group were given the same amount of distilled water.The general conditions of rats in each group were observed and their body mass was recorded.The changes of electromyography were observed.The levels of serum creatine kinase(CK),lactate dehydrogenase(LDH)and aspartate aminotransferase(AST)were detected by enzyme linked immunosorbent assay(ELISA).The weights of spleen and thymus of rats in each group were weighed,and the spleen index and thymus index were calculated.The pathological changes of skeletal muscle were observed by HE staining.The pathological injury of rat skeletal muscle was evaluated.The mRNA and protein expressions of interferon regulatory factor 3(IRF-3)and interferon regulatory factor 7(IRF-7)in rat skeletal muscle were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western blotting.Results Compared with the model group,the body mass,exercise unit time limit,wave amplitude,spleen index,thymus index,IRF-3 and IRF-7 mRNA and protein expression levels were increased,and the pathological scores of multiple waves,CK,LDH,AST and skeletal muscle of rats in low,medium and high dosage of PCP groups were decreased(P<0.05).Compared with the low dose of PCP group,the body mass,exercise unit time limit,wave amplitude,spleen index,thymus index,IRF-3 and IRF-7 mRNA and protein expression levels were increased,and the pathological scores of multiple waves,CK,LDH,AST and skeletal muscle of rats in medium and high dose of PCP groups were decreased(P<0.05).The changes of various indexes in the high dose group were more significant than those in the medium dose group(P<0.05).Conclusion PCP can regulate immune function and improve muscle injury in IIM rats,which may play a role by activating the expression of IRF-3 and IRF-7.
