Screening of anti-inflammatory fractions and chemical constituents from Potentilla kleiniana Wight.et Arn.
Objective To screen the anti-inflammatory fractions of Potentilla kleiniana Wight.et Arn.and to study the chemical constituents from the active fractions.Methods The anti-inflammatory activity of water fraction(FPA),50%ethanol fraction(FPB)and 95%ethanol fraction(FPC)of 60%ethanol extract Potentilla kleiniana Wight.et Arn.were evaluated by mice ear swelling acute inflammation model induced by xylene.The constituents from active fraction were further isolated by MCI,Sephadex LH-20,silica gel,octadecyl-silylated silica gel column chromatography and other methods,and their chemical structures were elucidated on the basis of physicochemical properties and spectral analysis.Results The 50%ethanol fraction exhibited anti-inflammatory activity,and 10 compounds were isolated and their structures were identified as:psoralen(1),bis(2'-ethylheptyl)phythalate(2),toddalolactone(3),fuzitine(4),1,2-oxyanthraquinone(5),arbutin(6),kaemperol(7),tangeretin(8),isorhamnetin(9),and acacetin-7-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(10).Conclusion The 50%ethanol fraction is the anti-inflammatory fraction,from which 10 compounds were separated and elucidated.Compouds 2-4,7-10 were isolated from Potentilla genus for the first time,and compounds 1 and 6 were isolated from Potentilla kleiniana Wight.et Arn.for the first time.
