Study on the action mechanism of Aquilariae lignum Resinatum in sleep disorders and anxiety diseases based on network pharmacology
Objective To explore the action mechanism of Aquilariae lignum Resinatum in sleep disorders and anxiety disorders by using network pharmacology.Methods The data of small molecule compounds corresponding to Aquilariae lignum Resinatum were searched and downloaded through Tcmsp and Herb databases,then the target genes corresponding to small molecule compounds and sleep disorders and anxiety were searched from Herb databases,and a drug-component-target-disease network was constructed with Cytoscape.Based on Gene Ontology and KEGG Pathway Database of biochemical pathways,the candidate genes were analyzed for function enrichment,and the protein-protein interaction(PPI)was analyzed by using STRING.Results 112 small molecule compounds of Aquilariae lignum Resinatum were screened,and their corresponding targets were obtained.831 corresponding genes of sleep disorder and anxiety were searched in Herb database,and 82 key targets of components and diseases were obtained.Among them,zoomaric acid,6,7-dimethoxy-2-(27-phenylethyl)chromone and acetic acid may be the important components of Aquilariae lignum Resinatum in the treatment of sleep disorders and anxiety;SRC,CTNNB1,TNF,PPARG,FOS and other genes may be important potential targets of Aquilariae lignum Resinatum in the treatment of sleep disorders and anxiety.GO analysis showed the target involved 1 279 biological process items,97 cell component items and 123 molecular function items.116 pathways was enriched by using KEGG,mainly involving cancer pathway,dopaminergic synapse,lipid and atherosclerosis,breast cancer and other key pathways.Conclusion Zoomaric acid,6,7-dimethoxy-2-(27 phenylethyl)chromone,acetic acid may be the components for the treatment of anxiety and sleep disorders,and the active components may play a role in the treatment of anxiety and sleep disorders by acting on SRC,CTNNB1,TNF,PPARG,and FOS.
