Study on the mechanism of resveratrol on pancreatic acinus apoptosis in rats with severe acute pancreatitis based on NOD like receptor pyrin domain containing 3 inflammatory body axis
Objective To investigate the effect of resveratrol on apoptosis of pancreatic acinar cells in rats with severe acute pancreatitis(SAP)and modulating the inflammatory body axis of NOD like receptor pyrin domain containing 3(NLRP3).Methods 50 rats were randomly divided into sham operation group,model group,low-dose resveratrol group,high-dose resveratrol group and dexamethasone group,with 10 rats in each group.Except for the sham operation group,the SAP model was prepared by retrograde injection of sodium bezoar cholate in other groups.5 minutes after the operation,the low-dose and high-dose resveratrol groups were intraperitoneally injected with 30 and 60 mg·kg-1 resveratrol,the dexamethasone group was intraperitoneally injected with 2 mg·kg-1 dexamethasone,once a day for consecutive 3 days,and the sham operation group and model group were intraperitoneally injected with the same amount of normal saline.The contents of amylase,lipase,interleukin(IL)-1β,IL-18 and tumor necrosis factor α(TNF-α)in serum of rats were detected,The pathological damage of pancreatic tissue was detected and recorded in histopathological score,and the apoptosis index of pancreatic acinar cells was detected.The protein relative expression levels of NLRP3,aspartic acid specific cysteine protease 1(caspase-1),B-cell lymphoma-2(Bcl-2)and Bcl-2 associated X protein(Bax)were also detected.Results In sham operation group,the pancreas tissue structure was normal without edema,hyperemia or necrosis.In model group,pancreatic interstitial edema was observed,alveolar septum widened,a large number of inflammatory cell infiltration,and punctate or lamellar hemorrhage or necrosis occurred in the parenchyma.The degree of pancreatic tissue lesion was reduced in the low-dose,high-dose resveratrol groups and dexamethasone group.Compared with sham operation group,the contents of amylase and lipase,IL-1β,IL-18 and TNF-α,histopathological score,apoptosis index and the relative expression levels of NLRP3,caspase-1 and Bax proteins in model group were increased,while the relative expression level of Bcl-2 protein was decreased(P<0.05).Compared with model group,the contents of amylase and lipase,IL-1β,IL-18 and TNF-α,histopathological score,apoptosis index and the relative expression levels of NLRP3,caspase-1 and Bax proteins in resveratrol low-dose,high-dose groups and dexamethasone group were decreased,while the relative expression level of Bcl-2 protein was increased(P<0.05).Compared with the low-dose resveratrol group,the contents of amylase and lipase,the contents of IL-1β,IL-18 and TNF-α,histopathological score,apoptosis index and the relative expression levels of NLRP3,caspase-1 and Bax proteins in the high-dose resveratrol group and dexamethasone group were decreased,while the relative expression level of Bcl-2 protein was increased(P<0.05).Compared with resveratrol high-dose group,the contents of amylase and lipase,IL-1β,IL-18 and TNF-α,histopathological score,apoptosis index and the relative expression levels of NLRP3,caspase-1 and Bax proteins in dexamethasone group were decreased,while the relative expression level of Bcl-2 protein was increased(P<0.05).Conclusion Resveratrol can reduce pancreatic injury and inhibit pancreatic acinocyte apoptosis in SAP rats,possibly by blocking the activation of NLRP3 inflammatory body axis.
