Mechanism of berberine in treating peri-implant inflammation in rats
Objective To investigate the effect of berberine in the treatment of peri-implantitis in rats and its effect on Toll-like receptor 4(TLR4)/nuclear factor κB(NF-κB)signaling pathway.Methods SD rats were used to build the peri-implantitis model,and randomly divided into model group,low-dose and high-dose berberine groups and antibiotic group,and a negative control group was set up.The low-dose and high-dose berberine groups were given 100 and 200 mg·kg-1 berberine,the antibiotic group was given 200 mg·kg-1 Amoxicillin Capsules and 100 mg·kg-1 Metronidazole Tablets,the model group and the negative control group were given equal volume of normal saline,continuous intervention for 4 weeks.The general conditions of the implants in each group were observed,the probing depth(PD)and gingival index(GI)of the implants were measured with a periodontal probe.The bone height and bone mineral density around the implants were measured by computed tomography(CT).The contents of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in peri-implant gingival tissue were detected by enzyme linked immunosorbent assy(ELISA).The TLR4/NF-κB signaling pathway-related protein expression in peri-implant gingival tissue of rats were detected by Western blotting.Results In the model,low-dose berberine,high-dose berberine,antibiotic and negative control groups,the PD and GI indexes of the rats gradually decreased,the bone height and bone density around the implant gradually increased,and the levels of TNF-α and IL-1β in the gingival tissue gradually decreased,and the activities of collagen-Ⅱ(COL-Ⅱ)and matrix metalloproteinase-13(MMP-13)in the gingival crevicular fluid of the implant gradually increased(P<0.05).In the model,the low-dose berberine,high-dose berberine and negative control group,the relative expressions of TLR4,MyD88 and p-NF-κB-65 proteins in the gingival tissue around the implants of the rats gradually decreased(P<0.05).Conclusion Berberine has a certain therapeutic effect on peri-implantitis in rats,and its mechanism may be related to the inhibition of TLR4/NF-κB signaling pathway.
