Effects of berberine on blood lipid metabolism and insulin resistance in diabetic rats
Objective To investigate the effect of berberine on blood lipid metabolism and insulin resistance(IR)in diabetes mellitus(DM)rats.Methods Ten normal rats were taken as the normal control group(Con).The rats with successful DM modeling were randomly divided into DM model group(M),positive control group(Pon),berberine low(L-Ber)and high-dose groups(H-Ber),which were 9,9,9 and 8 respectively.The other 8 non-DM rats were included in the non-DM group(N-DM).After the corresponding intervention,the body weight,drinking water,food intake,blood glucose,blood lipid and IR were measured.The histomorphological changes of pancreas were observed by HE staining.The protein expressions of phosphorylated insulin receptor substrate 1(p-IRS-1),the protein expression of phosphorylated phosphatidylinositol 3-kinase(p-PI3K)and glucose transporter-4(GLUT-4)were detected by Western blotting.Results Compared with the Con group,the water intake,food intake,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),fasting insulin,(FIns),homeostasis model assessment of insulin resistance,(HOMA-IR),glucose tolerance,fasting blood glucose(FBG)increased,the body weight,high density lipoprotein cholesterol(HDL-C)and the relative expression of p-IRS-1,p-PI3K and GLUT-4 protein decreased(P<0.05).Compared with the M group,the water intake,food intake,TC,TG,LDL-C,FIns,HOMA-IR,glucose tolerance,and FBG in the L-Ber group,H-ber group,and Pon group decreased in sequence,the abnormal morphology of the pancreas were reduced,the body weight,HDL-C and the relative expression of p-IRS-1,p-PI3K and GLUT-4 protein increased successively(P<0.05).Conclusion Berberine can improve blood lipid metabolism and IR in DM rats.The mechanism may be related to the regulation of IRS-1/PI3K/GLUT4 pathway.
