盐酸普拉克索药剂学的研究进展

摘要
目的 了解盐酸普拉克索的药动学及药剂学方面的研究进展,为盐酸普拉克索新剂型的进一步开发提供参考以及新思路.方法 通过查阅文献,对盐酸普拉克索的药动学及药剂学研究进行综述.结果 盐酸普拉克索是选择性非麦角类多巴胺受体激动剂,一般用于帕金森病和不宁腿综合征的治疗,同时还具有多种药理作用,如抗抑郁作用、神经元保护作用、抗氧化作用和抗炎作用,受到医药界的广泛关注和重视.盐酸普拉克索具有口服吸收迅速、生物利用度高等特点.目前以盐酸普拉克索作为单一主药上市的制剂为口服途径给药的普通片剂与缓释片剂,但需多次服用药物,易出现血药浓度波动的现象,因此患者顺应性差,亟需得到改善.结论 缓控迟释制剂、经皮给药制剂近年来发展迅速,逐步成为盐酸普拉克索制剂研发的重点.
Abstract
Objective In order to understand the advances of pharmacokinetics and pharmaceutical aspects of pramipexole dihydrochloride,and provide a reference for research and development.Methods The pharmacokinetics and pharmaceutical aspects of pramipexole dihydrochloride were reviewed by consulting published articles.Results Pramipexole dihydrochloride is a selective no-nergot dopamine receptor agonist,which is generally used in the treatment of Parkinson's disease and restless leg syndrome.At the same time,it also has a variety of pharmacological effects,such as antidepressant effect,neuroprotective effect,antioxidant effect and anti-inflammatory effect,which has been widely studied and valued by the pharmaceutical industry.Pramipexole dihydrochloride has the characteristics of rapid oral absorption and high bioavailability.At present,pramipexole hydrochloride is used as a single main drug listed preparation for oral administration of ordinary tablets and sustained-release tablets.The drug should be taken many times,which is prone to the fluctuations in blood concentration.Therefore,patients have poor compliance and need to be improved urgently.Conclusion Sustained-release,controlled-release drugs and transdermal drug delivery drugs have been developed rapidly in recent years,and have gradually become a focus of pramipexole dihydrochloride preparations.
