摘要
目的 初步阐明抗类风湿性关节炎方剂通痹化瘀膏的药效物质基础,为其临床应用提供科学依据.方法 用硅胶柱色谱、反相硅胶柱色谱、高效液相色谱等现代分离技术对通痹化瘀膏的有效部位进行分离纯化,结合化合物的理化性质和各种波谱技术对分离得到的化合物进行结构鉴定.结果 从通痹化瘀膏有效部位中分离鉴定出 14 个化合物,分别是花椒素(1)、2-methoxyfuranoguaia-9-ene-8-one(2)、棕榈酸(3)、2-methoxy-5-acetoxy fruranogermacr-1(10)-en-6-one(4)、sarcophytol M(5)、n-tetradecanyl oleate(6)、alismol(7)、2-methoxy-8,12-epoxygermacra-1(10),7,11-trien-6-one(8)、bis(2-ethyloctyl)phthalate(9)、(24S)-24-ethylcholest-5-en-3β-ol(10)、myrrhterpenoid E(11)、okilactomycin D(12)、bis(2-ethylhexyl)benzene-1,2-dicarboxylate(13)和5-羟甲基糠醛(14).结论 这14个化合物包括7个萜类、3个芳香族化合物、2个脂肪酸或酯、1个甾体和1个糠醛.本研究是首次对院内制剂通痹化瘀膏的药效物质基础进行研究.
Abstract
Objective To preliminarily study the effective substance of the active part of Tongbihuayu Ointment and to provide scien-tific evidence for its clinic application.Methods The active part of Tongbihuayu Ointment was purified by silica gel column chroma-tography,ODS column chromatography,HPLC and other methods.And the structures were characterized by spectroscopic analysis on the basis of their physicochemical properties.Results Fourteen compounds were isolated and identified as 1-(2-hydroxy-4,6-dime-thoxyphenyl)ethan-1-one(1),2-methoxyfuranoguaia-9-ene-8-one(2),hexadecanoic acid(3),2-methoxy-5-acetoxy fruranoger-macr-1(10)-en-6-one(4),sarcophytol M(5),n-tetradecanyl oleate(6),alismol(7),2-methoxy-8,12-epoxygermacra-1(10),7,11-trien-6-one(8),bis(2-ethyloctyl)phthalate(9),(24S)-24-ethylcholest-5-en-3β-ol(10),myrrhterpenoid E(11),okilactomy-cin D(12),bis(2-ethylhexyl)benzene-1,2-dicarboxylate(13)and 5-(hydroxymethyl)furan-2-carbaldehyde(14).Conclusion These compounds included 7 terpenoids,3 phenyl compounds,2 fatty acids or esters,1 steroid and 1 furaldehyde.This is the first time to study the effective substance of Tongbihuayu Ointment.
