银马解毒颗粒调控急性肺损伤小鼠肺部炎症的效果

目的 分析银马解毒颗粒调控白化家鼠亚系(laboratory-bred strain,BALB/c)急性肺损伤小鼠肺部炎症的效果及安全性。方法 选取 6~8周龄C57BL/6J小鼠,用脂多糖(lipopolysaccharide,LPS)成功构建 70只BALB/c急性肺损伤小鼠肺部炎症模型,并选取10只6~8周龄C57BL/6J小鼠作为空白组A。将70只模型小鼠随机分为7组(模型组B、银马解毒颗粒低剂量预防给药组C1、银马解毒颗粒高剂量预防给药组C2、银马解毒颗粒低剂量治疗组D1、银马解毒颗粒高剂量治疗组D2、银马解毒颗粒低剂量持续给药组E1和银马解毒颗粒高剂量持续给药组E2),每组10只。比较各组小鼠的药物毒性情况、肺泡灌洗液细胞计数,炎症因子和肺组织伊红染色法(hematoxylin-eosin staining,HE)染色情况。结果 造模过程中共 6只小鼠死亡,造模成功率为92。11%。与对照组比较,模型组的肺泡灌洗液炎症细胞数、炎症因子和HE染色炎症细胞浸润率均升高(P<0。05)。与正常肺组织比较,急性肺损伤模型鼠的肺组织HE染色可见肺泡间隔充血、水肿以及大量炎性细胞的浸润。成功造模的小鼠肺泡灌洗液细胞计数和炎症因子水平均高于A组,差异有统计学意义(P<0。05)。模型组小鼠的状态较差,表现为毛发耸立、抱团,进食量与饮水量明显减少,体质量减轻,大便干燥。各组急性肺损伤模型小鼠给予银马解毒颗粒灌胃1 d后,C2组、D2组和E2组小鼠的状态均较佳。造模8 d,模型组(B组)小鼠的死亡率达30%,显著高于其他各组(P<0。05);病理学上,与正常肺组织比较,急性肺损伤模型鼠的肺组织HE染色可见肺泡间隔充血、水肿以及大量炎性细胞的浸润。给予银马解毒颗粒后,肺泡间隔充血、水肿以及炎性细胞的浸润明显减少,且上述效果与银马解毒颗粒的剂量呈正比。在肺泡灌洗液细胞数方面,与A组比较,B组小鼠白细胞、中性粒细胞和淋巴细胞计数均明显增加。与E组和B组小鼠比较,给药组C组和D组3类细胞计数均显著降低(P<0。05);且C2组、D2组和E2组小鼠的3类细胞计数分别低于C1组、D1组和E1组(P<0。05)。在炎症因子方面,与A组比较,B组IL-6、TNF-α和IL-1β的质量浓度均显著增加(P<0。05)。与B组比较,C1、D1、E1组小鼠的IL-1β的质量浓度均降低,而C2组、D2组和E2组小鼠的TNF-α和IL-1β的质量浓度均降低(P<0。05);且C2组、D2组和E2组小鼠的IL-6、TNF-α和IL-1β的质量浓度分别低于C1组、D1组和E1组(P<0。05)。结论 银马解毒颗粒对于BALB/c急性肺损伤有显著保护性作用,能够改善其肺部炎症细胞及炎症因子的激活情况,从而缓解急性肺损伤症状。
Effect of Yinma Jiedu Granules on regulating lung inflammation in acute lung injury mice
Objective To analyze the effect and safety of Yinma Jiedu Granules on regulating lung inflammation in laboratory-bred strain(BALB/c)acute lung injury mice.Methods 70 C57BL/6J male mice of 6-8 weeks old were selected to construct BALB/c acute lung injury inflammation model by using lipopolysaccharide(LPS),and the other 10 C57BL/6J male mice of 6-8 weeks old were used as blank group A for the control.The 70 model mice were randomly divided into 7 groups(model group B,low-dose prophylactic treatment group C1 of Yinma Jiedu Granules,high-dose prophylactic treatment group C2 of Yinma Jiedu Granules,low-dose treatment group D1 of Yinma Jiedu Granules,high-dose treatment group D2 of Yinma Jiedu Granules,low-dose continuous treatment group E1 of Yinma Jiedu Granules,and high-dose continuous treatment group E2 of Yinma Jiedu Granules),with 10 mice in each group.The drug toxicity,alveolar lavage fluid cell count,inflammatory factors and hematoxylin-eosin(HE)staining of lung tissue in each group were compared.Results During the modeling process,a total of 6 mice died,with a success rate of 92.11%.Compared with the control group,the number of inflammatory cells,inflammatory factors,and HE staining infiltration rate of inflammatory cells in the alveolar lavage fluid of the model group were all increased(P<0.05).Compared with normal lung tissue,HE staining of lung tissue in acute lung injury model mice showed alveolar septal congestion,edema,and infiltration of a large number of inflammatory cells.The cell count and inflammatory cytokine levels in the alveolar lavage fluid of successfully modeled mice were higher than those in Group A,and the differences were statistically significant(P<0.05).The state of the model group mice was poor,characterized by towering and clumping hair,significantly reduced food and water intake,reduced body weight,and dry stools.After administration of Yinma Jiedu Granules orally for 1 day to each group of acute lung injury model mice,the C2,D2,and E2 groups of mice were in good condition.After 8 days of modeling,the mortality rate of model group(Group B)mice reached 30%,significantly higher than other groups(P<0.05);Pathologically,compared with normal lung tissue,the HE staining of lung tissue in acute lung injury model mice showed congestion and edema of alveolar septa,as well as infiltration of a large number of inflammatory cells.After administration of Yinma Jiedu Granules,the congestion,edema,and infiltration of inflammatory cells in the alveolar septa were significantly reduced,and the above effects were proportional to the dosage of Yinma Jiedu Granules.In terms of cell count in alveolar lavage fluid,compared with group A,group B mice showed a significant increase in white blood cell,neutrophil,and lymphocyte counts.Compared with mice in Group E and Group B,the 3 types of cell counts in Group C and Group D of the treatment group were significantly reduced(P<0.05);And the counts of 3 types of cells in the C2,D2,and E2 groups of mice were lower than those in the C1,D1,and E1 groups,respectively(P<0.05).In terms of inflammatory factors,compared with Group A,Group B showed the levels of IL-6,TNF-α and IL-1β all significantly increased(P<0.05).Compared with Group B,the levels of IL-1β in C1,D1,and E1 groups decreased,while the levels of TNF-α and IL-1β in the C2,D2,and E2 groups of mice decreased(P<0.05);And the IL-6,TNF-α and IL-1β in C2,D2,and E2 groups of mice were lower than those in C1 group,D1 group,and E1 group(P<0.05).Conclusion Yinma Jiedu Granules have a significant protective effect on BALB/c acute lung injury,and can reduce the activation of inflammatory cells and inflammatory factors in lungs,thereby alleviate the symptoms of acute lung injury.
