金钱草提取物对胃癌大鼠化疗肾损伤的作用及机制

目的 探究金钱草提取物对胃癌大鼠化疗肾损伤的作用,并探讨可能的机制。方法 取50只SD大鼠建立胃癌模型,将建模成功的40只随机选10只设为胃癌组,其余建立化疗肾损伤模型,随机分为损伤组、干预组、金钱草联合核因子E2相关因子2(nuclear factor-E2 related factor2,Nrf2)抑制剂(ML385)组,各10只。干预组灌胃金钱草提取物800 mg·kg-1,金钱草联合ML385组灌胃金钱草提取物800 mg·kg-1及ML385 20 mg·kg-1,胃癌组和损伤组灌胃等体积生理盐水。每日1次,持续处理1周。检测血清尿素氮(blood urea nitrogen,BUN)、血清胱抑素 C(cystatinC,CysC)水平;计算肾脏质量指数;检测肾组织丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)水平,肾组织Nrf2、血红素加氧酶-1(heme oxygenase-1,HO-1)蛋白水平。结果 与损伤组比较,干预组的CysC、BUN水平、肾脏质量指数、MDA水平均降低,GSH-Px的活性增强(P<0。05);与干预组比较,金钱草联合ML385组的CysC、BUN水平、肾脏质量指数、MDA水平均增高,GSH-Px的活性减弱(P<0。05);与损伤组比较,干预组的Nrf2、HO-1蛋白水平均增高(P<0。05);与干预组比较,金钱草联合ML385组的Nrf2、HO-1蛋白水平均降低(P<0。05)。结论 金钱草提取物可减轻胃癌化疗大鼠肾组织氧化应激,改善肾功能,可能是通过增强Nrf2/HO-1通路活性发挥作用的。
Effects and mechanisms of the extract of Lysimachia chinensis L.on renal injury caused by chemotherapy in rats with gastric cancer
Objective To explore the effect of the extract of Lysimachia chinensis L.(ELC)on renal injury in rats with gastric cancer after chemotherapy,and to explore the possible mechanism.Methods Fifty SD rats were selected to establish gastric cancer model,and 10 of the 40 successfully modeled rats were randomly selected as gastric cancer group,and the rest were randomly divided into injury group,intervention group and ELC combined with nuclear factor-E2 related factor2(Nrf2)inhibitor(ML385)group,with 10 rats in each.The intervention group was administered with 800 mg·kg-1 of ELC,the ELC combined with ML385 group was administered with 800 mg·kg-1 of ELC and 20 mg·kg-1 of ML385,the gastric cancer group and the injury group were administered with equal volume of normal saline.Once a day for 1 week.The serum urea nitrogen(BUN)and serum creatinine(CysC)levels were detected.The kidney mass index was calculated.The malondialdehyde(MDA),superoxide dismutase(GSH-Px)levels,the protein expression of nuclear factor E2 related factor 2(Nrf2)and heme oxygenase 1(HO-1)in kidney tissue were detected.Results Compared with the injury group,the levels of CysC,BUN,kidney mass index,and MDA in the intervention group were decreased,of which the activities of GSH-Px were increased(P<0.05).Compared with the intervention group,the levels of CysC,BUN,kidney mass index,and MDA in the ELC combined with ML385 group were increased,of which the activities of GSH-Px were decreased(P<0.05).Compared with the injury group,the expression of Nrf2 and HO-1 protein in the intervention group were increased(P<0.05).Compared with the intervention group,the expression of Nrf2 and HO-1 protein in the ELC combined with ML385 group were decreased(P<0.05).Conclusion The extract of ELC may act through activating the Nrf2/HO-1 signaling pathway to reduce the oxidative stress and kidney damage in the kidney tissue of rats with gastric cancer chemotherapy,and to protect the renal function.
