Effect of Asarum Decoction on analgesia and birth process in labor rats
Objective To observe the effect of Asarum Decoction(AG)on the analgesia and birth process in labor rats,and to explore the possible mechanism.Methods Forty pregnant rats were randomly divided into normal group,AG group,phorbol ester group,AG combined with phorbol ester group,with 10 in each group.On the 18th day of conception,the AG combined with phorbol ester group was given gastric AG 12.0 g·kg-1,and 1 μg phorbol ester was injected subcutaneously.In the phorbol ester group,the same amount of distilled water was given by gavage,and 1 μg phorbol ester was injected subcutaneously.The AG group was given gastric AG 12.0 g·kg-1,and an equal volume of normal saline was injected subcutaneously.The normal group was given the same volume of distilled water by intragastric administration;the same volume of normal saline was subcutaneously injected.The interventions were once a day for 3 days.The labor time was recorded.Hot water flick method was used to detect the pain threshold.The levels of serum 5-hydroxytryptamine(5-HT),dynorphin(DYN)and glutathione(GSH)were tested.Western blotting was used to detect the protein expression of spinal cord tissue protein kinase Cε(PKCε)and transient receptor potential vanilloid receptor 1(TRPV1).Results Compared with in normal group,the labor start time,half-labor time and total labor time were shortened,the pain threshold,serum DYN and GSH levels were increased,and the serum 5-HT levels,spinal cord tissue PKCε and TRPV1 protein expression were decreased in AG group(P<0.05).The labor start time,half labor time and total labor time were prolonged,the pain threshold,serum DYN and GSH levels were decreased,the serum 5-HT levels,spinal cord tissue PKCε and TRPV1 protein expression were increased in phorbol ester group(P<0.05).Compared with the AG combined with phorbol ester group,the onset time of labor,the half-time of labor and the total labor time in the AG group were shortened,the pain threshold,serum DYN and GSH levels were increased,and the serum 5-HT level and the expressions of PKCε and TRPV1 protein in spinal cord tissue were decreased(P<0.05).The labor onset time,half-partum time and total labor time were prolonged,the pain threshold,serum DYN and GSH levels decreased,the serum 5-HT level,spinal cord tissue PKCε and TRPV1 protein expressions in the phorbol ester group were increased(P<0.05).Conclusion AG can shorten the labor process and exert analgesic effect,and its mechanism of action may be related to the inhibition of PKCε/TRPV1 pathway.
