Effect and mechanism of ligustrazine on fetal development in rats with experimental surgical fetal growth restriction
Objective To investigate the effects of ligustrazine(tetramethyl pyrazine)on fetal development in experimentally operated fetal growth restriction(FGR)rats,and the effect on the expression levels of soluble vascular endothelial growth factor receptor-1(sFlt-1)and placental growth factor(PLGF)in placental tissue of rats.Methods Twenty-four pregnant rats were randomly divided into normal group,model group,and ligustrazine group(80 mg·kg-1),with 8 rats in each group.Except for the normal group,the other 2 groups were started to establish the FGR rat model by smoking method on the 5th day of pregnancy.On the 21st day of pregnancy,cesarean section was performed to evaluate fetal outcomes,to check fetal growth and development,and to detect the mRNA and protein expressions of sFlt-1 and PLGF in placental tissue of rats.Results Compared with the normal group,the embryo absorption rate,the expression level of sFlt-1 mRNA and protein in rat placenta,the abnormal rate and variation rate of internal organs,and the abnormal rate and variation rate of bones in the model group were significantly increased(P<0.05).The expression level of PLGF mRNA and protein in rat placenta,the number of surviving fetuses,the quality of living carcass,and the ossification rate of metacarpal,metatarsal and coccygeal vertebrae decreased significantly(P<0.05).Compared with the model group,the embryo absorption rate,the expression level of sFlt-1 mRNA and protein in rat placenta,the abnormal rate and variation rate of visceral organs,and the abnormal rate and variation rate of bones in ligustrazine group were significantly decreased(P<0.05).The expression level of PLGF mRNA and protein in rat placenta,the number of surviving fetuses,the quality of living carcass,and the ossification rate of metacarpal,metatarsal and coccygeal vertebrae increased significantly(P<0.05).Conclusion Ligustrazine can significantly down-regulate the expression of sFlt-1 and up-regulate the expression of PLGF in FGR pregnant rats,promote fetal growth and development,and improve fetal growth restriction.
