Effect of Longshengzhi Capsules on oxidative stress damage in human umbilical vein endothelial cells
Objective To investigate the effect of Longshengzhi Capsules on oxidative stress injury of human umbilical vein endothelial cells(HUVEC).Methods A hydrogen peroxide endothelial cell injury model was established,and different doses of Longshengzhi Capsules ethanol extract(2.5,5.0 and 10.0 mg·mL-1)and simvastatin(5 mg·mL-1)were administered respectively.CCK8 method was used to detect the cell activity,and enzyme-linked immunosorbent assay(ELISA)was used to detect the inflammation-related factors[interleukin(IL)-1β、IL-6、IL-12 and tumor necrosis factor-α(TNF-α)]in the cell supernatant.Nitric oxide(NO)was detected by nitric reductase method,and reactive oxygen species(ROS)were detected by flow cytometry.Results Compared with the blank group,the cell viability and NO content in the model group were significantly decreased.The levels of inflammation-related factors(IL-1β,IL-6,IL-12 and TNF-α)and ROS were significantly increased.After the intervention of Longshengzhi Capsules and simvastatin,the above indexes were reversed in all groups(P<0.05),and there was a certain correlation with the dose.Conclusion Longshengzhi Capsules can protect HUVEC induced by hydrogen peroxide by increasing cell activity,inhibiting inflammation and oxidative stress reaction.
