目的 探讨龙生蛭胶囊对人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVEC)氧化应激损伤的影响。方法 建立过氧化氢内皮细胞损伤模型,给予不同剂量的龙生蛭胶囊乙醇提取物(2。5、5。0、10。0 mg·mL-1)和辛伐他汀(5 mg·mL-1)干预。采用CCK8法检测细胞活性,用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测细胞上清液的炎症相关因子[白细胞介素(interleukin,IL)-1β、IL-6、IL-12和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)]水平,用硝酸还原酶法检测一氧化氮(nitric oxide,NO)含量,用流式细胞仪检测活性氧(reactive oxygen species,ROS)水平。结果 与空白组相比,模型组中细胞活力和NO含量均显著下降,而炎症相关因子(IL-1β、IL-6、IL-12和TNF-α)和ROS水平明显上升,给予龙生蛭胶囊和辛伐他汀干预后,上述各组指标均呈现逆转(P<0。05),且与剂量呈一定相关性。结论 龙生蛭胶囊通过提高细胞活性,抑制炎症和氧化应激反应,从而对由过氧化氢诱导的HUVEC氧化应激损伤发挥保护作用。
Effect of Longshengzhi Capsules on oxidative stress damage in human umbilical vein endothelial cells
Objective To investigate the effect of Longshengzhi Capsules on oxidative stress injury of human umbilical vein endothelial cells(HUVEC).Methods A hydrogen peroxide endothelial cell injury model was established,and different doses of Longshengzhi Capsules ethanol extract(2.5,5.0 and 10.0 mg·mL-1)and simvastatin(5 mg·mL-1)were administered respectively.CCK8 method was used to detect the cell activity,and enzyme-linked immunosorbent assay(ELISA)was used to detect the inflammation-related factors[interleukin(IL)-1β、IL-6、IL-12 and tumor necrosis factor-α(TNF-α)]in the cell supernatant.Nitric oxide(NO)was detected by nitric reductase method,and reactive oxygen species(ROS)were detected by flow cytometry.Results Compared with the blank group,the cell viability and NO content in the model group were significantly decreased.The levels of inflammation-related factors(IL-1β,IL-6,IL-12 and TNF-α)and ROS were significantly increased.After the intervention of Longshengzhi Capsules and simvastatin,the above indexes were reversed in all groups(P<0.05),and there was a certain correlation with the dose.Conclusion Longshengzhi Capsules can protect HUVEC induced by hydrogen peroxide by increasing cell activity,inhibiting inflammation and oxidative stress reaction.
