为了研究黑果枸杞多糖的最佳提取工艺条件和枸杞多糖抗肿瘤活性,通过改变料液比、提取温度、提取时间等影响因素设计正交试验.提取的黑果枸杞多糖溶液经醇沉、抽滤、干燥等处理得粗多糖粉末,该粉末经大孔树脂脱色、Sevage法脱蛋白,精制得到黑果枸杞多糖.结果表明,黑果枸杞多糖的最佳提取工艺条件为:料液比1∶25,提取温度 80℃,提取时间 3 h,多糖提取率为11.58%;黑果枸杞多糖体外有抑制人慢性粒细胞白血病细胞株K562 增殖的作用,有望成为一种理想的抗肿瘤佐剂.
Extraction and Anti-tumor Activity Analysis of Polysaccharides from Lycium Ruthenicum
To study the optimal extraction process conditions and anti-tumor activity of polysaccharides from lycium ruthenicum,orthogonal experiments were designed by changing factors such as material liquid ratio,ex-traction temperature,and extraction time.The extracted lycium ruthenicum polysaccharide solution was treated with alcohol precipitation,filtration,and drying to obtain crude polysaccharide powder.The powder was decolor-ized by macroporous resin,deproteinized by Sevage method,and refined to obtain lycium ruthenicum polysaccha-ride.The results showed that the optimal extraction process conditions for polysaccharides from lycium rutheni-cum were material liquid ratio 1∶25,extraction temperature 80℃,extraction time 3 hours,and the polysaccha-ride extraction rate was 11.58%;Lycium ruthenicum polysaccharides have an inhibitory effect on the prolifera-tion of human chronic myeloid leukemia cell line K562 in vitro,and are expected to become an ideal anti-tumor adjuvant.
