Studies on the ability and activity of Escherichia coli and Staphylococcus aureus biofilm formation
Objective:To study the ability of Escherichia coli(E.coli)and Staphylo-coccus aureus(S.aureus)biofilm formation and the metabolic activity of bacteria in their bio-films.Methods:Standard strains of E.coli and S.aureus were used to determine their ability to form biofilms in different incubation times,mixing ratios,and pH cultures,and to determine the metabolic activity of the bacteria in their biofilms.Results:The biofilm formation ability of E.coli and S.aureus was higher at 48h than at 24h(P<0.05)and 72h(P>0.05).E.coli∶S.au-reus cultured according to the 1∶105 mixing ratio had the strongest biofilm formation ability,and the differences were statistically significant when compared with the rest of the mixing ratios and the culture of the two bacteria alone(P<0.05).The biofilm formation ability and the meta-bolic activity of bacteria in biofilm,measured at 48h of mixed culture of E.coli∶S.aureus ac-cording to 1∶105 in different pH cultures,were the highest at pH 8.0,and were stronger than those of E.coli(P<0.05)and S.aureus(P<0.05)cultured alone.The ability of biofilm for-mation and the metabolic activity of the bacteria in the biofilm of the mixed bacterial solution were lowest at pH 4.0.Conclusion:The overall structure of the bacterial aggregate periplasm formed when cultured according to E.coli∶S.aureus as a 1∶105 mixture for48h was the most stable,and on the basis of the stable structure,the growth and reproduction of the bacteria and their invasion ability were stronger when the pH of the TSB culture solution was 8.0.A weakly acidic environment reduces the biofilm-forming ability and activity of two common pathogens of AV when cultured alone and in combination.
