The expression and clinical significance of ADH1B and ALDH2 in gastric cancer
Objective To investigate the expression of alcohol dehydrogenase 1B(ADH1B)and aldehyde dehydrogenase 2(ALDH2)and their clinical significance in gastric cancer patient.Methods Totally,132 gastric cancer patients who underwent curative resection in Chuzhou First People's Hospital from May 2017 to October 2019 were included.The expression of ADH1B and ALDH2 in gastric cancer and adjacent non-cancer tissues were detected by immunohistochemistry(IHC).The relationships between the expression of ADH1 B and ALDH2 and clinicopathologic characteristics were analyzed,and their prognostic value for gastric cancer patients were evaluated using Kaplan-Meier curves.Univariate and multivariate Cox regression analysis were conducted to determine independent prognostic factors for gastric cancer patients.Results The IHC analysis of 132 tissue samples showed that the positive rates of ADH1B(41.5%vs 58.5%,x2=9.594,P=0.002)and ALDH2 expression(38.8%vs 61.2%,x2=16.716,P<0.001)in gastric cancer tissues were significantly lower than in adjacent non-cancer tissues.Compared with high expression group,low expression of ADH1B was frequently detected in T3-T4 stage(x2=5.572,P=0.018)and pTNM stage Ⅲ(x2=4.675,P=0.031)and had a higher rate of lymphovascular invasion(x2=4.566,P=0.033).Also,low expression of ALDH2 was frequently detected in poorly-differentiated adenocarcinoma(x2=4.261,P=0.039)and pTNM stage Ⅲ(x2=5.877,P=0.015),and had a higher incidence of lymph node metastasis(x 2=5.491,P=0.019)than its high expression.Survival analysis indicated that low expression of ADH1B and ALDH2 were associated with poor prognosis of gastric cancer patients.The 3-year overall survival(OS)rates of patients with low and high expression of ADH1B were 52.3%and 73.9%,respectively(x2=6.900,P=0.009).The 3-year OS rates of patients with low and high expression of ALDH2 were 49.8%and 74.4%,respectively(x 2=8.665,P=0.003).The expression of ADH1B(HR=2.115,95%CI:1.133-3.946,P=0.019)and ALDH2(HR=2.296,95%CI:1.207-4.367,P=0.011)were identified as independent prognostic factors for gastric cancer patients.Conclusion The expression of ADH1B and ALDH2 were decreased in gastric cancer tissues,and they might be useful biomarkers for predicting tumor progression and survival outcome.
