Expression and clinical significance of LncRNA HAND2-AS1 in serum of patients with cervical cancer
Objective To investigate the expression and clinical significance of long non-coding RNA(lncRNA)heart and neural crest derivatives express transcript 2 antisense sequence 1(HAND2-AS1)in ser-um of patients with cervical cancer.Methods A total of 50 patients with cervical cancer admitted to Maternal and Child Health Hospital of Jiaozuo City from January 2019 to June 2020 were selected as the cervical cancer group,and were divided into lncRNA HAND-AS1 high expression group and lncRNA HAND-AS1 low ex-pression group according to the median lncRNA HAND-AS1 expression value,with 25 cases in each group.During the same period,55 patients with cervical intraepithelial neoplasia(CIN)diagnosed were selected as CIN group and 50 healthy subjects were selected as healthy control group.Real-time fluorescence quantitative polymerase chain reaction and electrochemiluminescence were used to detect the expression of serum lncRNA HAND2-AS1,cancer antigen 125(CA125)and squamous cell carcinoma-associated antigen(SCC)in each group,and their relationship with clinical characteristics and prognosis was analyzed.The receiver operating characteristic(ROC)curve was drawn to analyze the clinical diagnostic efficacy of combined detection of ser-um lncRNA HAND2-AS1,CA125 and SCC in cervical cancer.Results The expression of serum lncRNA HAND2-AS1 in cervical cancer group was significantly lower than that in CIN group and healthy control group.The expression level of LncRNA HAND2-AS1 was significantly correlated with tumor size,cervical in-filtration depth,FIGO stage and lymph node metastasis in cervical cancer patients(P<0.05).The area under the ROC curve of lncRNA HAND2-AS1 in the diagnosis of cervical cancer was 0.861(95%CI 0.792-0.930).The area under the ROC curve of HAND2-AS1,CA125 and SCC in the diagnosis of cervical cancer was 0.912(95%CI 0.908-0.952).The diagnostic efficiency was significantly higher than that of single index detection,and the difference was statistically significant(P<0.05).The 3-year survival rate of patients in ln-cRNA HAND-AS1 high expression group was significantly higher than that in lncRNA HAND-AS1 low ex-pression group,and the difference was statistically significant(P<0.05).LncRNA HAND2-AS1,CA125,SCC,FIGO stage and lymph node metastasis were related to the prognosis of patients with cervical cancer,and the differences were statistically significant(P<0.05).FIGO stage and SCC were independent risk factors af-fecting the prognosis of patients with cervical cancer,and the differences were statistically significant(P<0.05).Conclusion The expression of LncRNA HAND2-AS1 in serum of patients with cervical cancer is low,which is obviously correlated with tumor size,depth of cervical invasion,FIGO stage and lymph node metastasis.Detection of serum lncRNA HAND2-AS1 has certain value in the diagnosis and prognosis evaluation of cervical cancer.
