Analysis of HLA-A,B and HPA Gene Polymorphism of Platelet Donors in Nanchang and Establishment of Typing Library
Objectives By analyzing the distribution characteristics of gene loci HLA-A,B,and HPA 1-6,10,15,and 21 from platelet donors through detection,this paper aims to strengthen the establishment of a comprehensive platelet donor database in Nanchang,thusensuring theeffectiveness and timeliness of clinical platelet transfusion and improving the efficacy of clinical blood transfusion.Methods The blood samples of 800 platelet donors in Nanchang were genotyped for HLA-A,B loci and HPA 1-6,10,15,21 loci through fluorescence quantitative PCR,and the gene frequency and genotype frequency were calculated.Results A total of 45 HLA-A and HLA-B alleles were detected at HLA-A and B loci,of which 16 were HLA-A alleles,and A2,A11 and A24 were with higher frequency.A total of 29 HLA-B alleles were detected,and B13,15,B40 and B46 had higher frequencies.HPA3 and HPA15 had higher mismatch rate at HPA 1-6,10,15,and 21 loci,and the mismatch rate was less than 10%.Conclusions The HPA-HLA gene polymorphism in the population of Nanchang exhibits similarities to that observed in other Han ethnic regions.We consider the gene databases of other Han ethnic regions to obtain computational results and the distribution frequency of ABO blood types in Nanchang,as a database comprising 7000 individuals would be necessary to better meet the demands of patients.
