Effect of electro-acupuncture on pain behavior and trigeminovascular system in nitroglycerine induced migraine mice
Objective It is to observe the effect of electro-acupuncture(EA)on pain behavior and trigeminovascular system in nitroglycerine induced migraine mice,and to clarify its internal mechanism.Methods Twenty male C57BL/6 mice were randomly divided into control group,model group,sham acupuncture group and EA group,with 5 mice in each group.The mice of all groups except for the control group were given nitroglycerin by intraperitoneal injection on Day 1,3,5,7,9,11,13 of experiment to establish models of chronic migraine.The sham acupuncture group and EA group were re-spectively treated with sham EA and EA at 30 min after each injection.The periorbital mechanical pain threshold,hindfoot mechanical pain threshold and thermal pain threshold of the mice were respectively detected before injection on the day of the experiment(day 0)and on days 2,4,6,8,10,12,and 14 of the experiment;on day 14 of the experiment,the ser-um levels of nitric oxide(NO)and calcitonin gene-related peptide(CGRP)were detected by ELISA,and the expression of c-Fos protein in trigeminal ganglion was detected by immunofluorescence and Western blot.Results On days 6,8,10,12,and 14,the periorbital mechanical stimulation response scores of mice in the model group were significantly higher than those of mice in the control group during the same period(all P<0.05);on days 10,12,and 14,the periorbital mechan-ical stimulation response scores of mice in the EA group were significantly lower than those of mice in the model group dur-ing the same period(all P<0.05),and the scores of mice in the EA group were significantly lower than those in the sham EA group on days 12 and 14 of the experiment(all P<0.05).On days8,10,12 and14 of the experiment,the hindfoot 50%withdrawal thresholds of mice in the model group were significantly lower than those of mice in the control group(all P<0.05),and the latency period of hindfoot withdrawal of thermal pain was significantly shorter than that in the control group(P<0.05);on days 10,12 and 14 of the experiment,the hindfoot 50%withdrawal thresholds of mice in the EA group were significantly higher,and the latency period was significantly longer than those in the model group(all P<0.05),and the hindfoot 50%withdrawal threshold of mice in the EA group was significantly higher,and the latency period was significantly longer than those in the sham EA group(all P<0.05).The serum levels of NO and CGRP and the aver-age optical density of positive expression of c-Fos and the relative expression of c-Fos protein in trigeminal ganglion of mice in the model group were significantly higher than those in the control group(all P<0.05),all these indexes in the EA group were significantly lower than those in the model group(all P<0.05).Conclusion Electroacupuncture can signifi-cantly improve the pain behavior of mice with nitroglycerine induced chronic migraine,which may be achieved by reducing the concentration of NO and CGRP in serum and the expression of trigeminal sensitising protein c-fos,so as to improve the vascular dysfunction of trigeminal nerve.
