Electro-acupuncture alleviates hemiplegic spasm after cerebral hemorrhage via up-regulating GluR2 expression in cervical enlargement of spinal cord in rats
Objective It is to study the effect of electro-acupuncture on the expression of glutamate receptor 2(GluR2)in cervical enlargement of spinal cord in rats with hemiplegic spasm after cerebral hemorrhage,and to explore the mecha-nism of electroacupuncture in relieving hemiplegic spasm symptoms after cerebral hemorrhage.Methods Forty-five healthy SD rats were randomly divided into 9 rats in the sham operation group,18 rats in the model group and 18 rats in the electro-acupuncture group.The rats in the model group and electroacupuncture group were injected with autologous tail blood into the right internal capsule by stereotactic device to establish models of cerebral hemorrhage,and the rats in the sham opera-tion group was not injected with autologous tail blood.After modeling,the rats of the electro-acupuncture group were stimu-lated with electro-acupuncture at Quchi and Zusanli acupoints,while the sham operation group and model group were not given any intervention.Behavioral tests[neurological status(assessed by Zea-longa score),muscle tension(assessed by modified Ashworth grading scale),and open field test]were performed in the rats on the 3rd and 7th days after surgery,and the expression of GluR2 in cervical enlargement of spinal cord was detected by Western blot and immunohistochemistry,respectively.Results Compared with the sham operation group on postoperative days 3 and 7,the Zea-longa scores and modified Ashworth scores of the left forelimb and left hindlimb of the rats in the model group were significantly increased(all P<0.05),the total distance traveled for locomotion was significantly shortened(P<0.05),the mean velocity was significantly slowed(P<0.05),the resting time was significantly increased(P<0.05),and the expression of GluR2 in cervical enlargement of spinal cord was significantly reduced(P<0.05).Compared with the model group,the Zea-longa scores and modified Ashworth scores of the left forelimb and left hindlimb of the rats in the electro-acupuncture group were significantly decreased(all P<0.05),the total distance traveled for locomotion was significantly increased(P<0.05),the mean velocity was significantly increased(P<0.05),the resting time was significantly decreased(P<0.05),and the expression of GluR2 in cervical enlargement of spinal cord was significantly increased(P<0.05).Conclusion Electro-acupuncture stimulation at"Quchi"and"Zusanli"can significantly improve the spastic paralysis symptoms and relieve the increase of muscle tension after cerebral hemorrhage in rats.Up-regulating GluR2 expression in in cervical enlargement of spinal cord may be one of the mechanisms by which electroacupuncture alleviates limb spasm after intracerebral hemor-rhage.
