清化饮对脾胃湿热证抑郁大鼠血清5-HT与海马组织的影响

目的 探究清化饮对脾胃湿热证抑郁大鼠血清5-羟色胺(5-HT)与海马组织的影响.方法 取30 只SPF级SD 雄性大鼠,随机选择其中5 只作为空白组,其余25 只采用高脂高糖饮食和慢性不可预知温和应激方法建立脾胃湿热证抑郁模型,将造模成功的大鼠随机分为模型组6 只、清化饮组6 只、舍曲林组 6 只、清化饮+舍曲林组 7 只.清化饮组给予清化饮 27.5 g/kg灌胃,舍曲林组给予舍曲林片5.21 mg/kg灌胃,清化饮+舍曲林组给予清化饮27.5 g/kg和舍曲林片5.21 mg/kg灌胃,模型组给予等体积的生理盐水灌胃,均1 次/d,连续灌胃28 d.末次灌胃结束后,应用强迫游泳实验评价大鼠的抑郁状态,记录大鼠灌胃前后的体重,ELISA法检测血清 5-HT水平,HE染色观察海马组织病理形态.结果 模型组大鼠的强迫游泳静止时间明显长于空白组(P<0.05),清化饮组、舍曲林组、清化饮+舍曲林组均明显短于模型组(P均<0.05);各组间大鼠体重比较差异无统计学意义(P>0.05),但清化饮组大鼠体重变化量明显低于舍曲林组(P<0.05);模型组大鼠血清5-HT水平与空白组比较差异无统计学意义(P>0.05),舍曲林组和清化饮+舍曲林组均明显高于模型组(P均<0.05),清化饮+舍曲林组明显高于清化饮组(P<0.05).模型组大鼠海马体积减小,CA2~CA3 区神经元细胞减少,沟回带厚度变薄;各药物组大鼠海马组织CA1~CA3 区神经元细胞紧密排列,细胞形态未见异常,沟回带厚度较模型组厚;其中清化饮组、清化饮+舍曲林组海马组织CA1~CA3 区神经元细胞紧密均匀排列优于舍曲林组.结论 清化饮可改善脾胃湿热证大鼠抑郁样行为,其机制可能与提高血清5-HT水平,恢复海马组织病理形态有关.
Effects of Qinghua Decoction on serum 5-HT and hippocampal pathology in depression rats with syndrome of spleen-stomach dampness heat
Objective It is to study the effect of Qinghua Decoction on serum 5-HT and hippocampal pathology in de-pression rats with syndrome of spleen-stomach dampness heat.Methods Thirty male SPF Rats were selected,in which 5 rats were randomly selected as blank group,and the remaining rats were fed with high-fat and high-sugar diet and stimula-ted with chronic unpredictable mild stress to establish depression model of syndrome of spleen-stomach dampness heat.The successfully modeled rats were randomly divided into model group(n =6),Qinghua Decoction group(n =6),sertraline group(n =6),and Qinghua Decoction +sertraline group(n =7).The Qinghua Decoction group was given Qinghua De-coction 27.5 g/kg by gavage,the sertraline group was given sertraline 5.21 mg/kg by gavage,and the Qinghua Decoction + sertraline group was given Qinghua Decoction 27.5 g/kg +sertraline 5.21 mg/kg by gavage,the model group was given the same volume of normal saline by gavage,all once daily,continuously treated for 28 days.At the end of intragastric admin-istration,the depression status of the rats was evaluated by forced swimming test,their body weight before and after gavage was recorded,the content of 5-HT in serum was detected by ELISA,and the pathological morphology of hippocampus was observed by HE staining.Results The resting time of forced swimming of rats in the model group was significantly longer than that in the blank group(P<0.05),and those in the Qinghua Decoction group,sertraline group,and Qinghua Decoc-tion +sertraline group were significantly shorter than that in the model group(all P<0.05);there was no statistically sig-nificant difference in the body weight of rats among every groups(all P>0.05),but the change of body weight of rats in the Qinghua Decoction group was significantly lower than that in the sertraline group(P<0.05);the difference in the ser-um level of 5-HT of rats was not statistically significant between the model group and blank group(P>0.05),while the serum level of 5-HT in the sertraline group and Qinghua Decoction +sertraline group were significantly higher than that of the model group(both P<0.05),and the Qinghua Decoction +sertraline group was significantly higher than the Qinghua Decoction group(P<0.05).The hippocampal volume of rats in the model group was decreased,the neuronal cells in the CA2-CA3 area were reduced,and the thickness of sulcus gyrus was thinned;the neuronal cells in CA1-CA3 area of hipp-ocampal tissues of rats in each treatment group were tightly arranged,the cellular morphology was not abnormal,and the thickness of sulcus gyrus was thicker than that of rats in the model group;the neuronal cells in CA1-CA3 area of hippocam-pal tissues were tightly and uniformly arranged in Qinghua Decoction group and Qinghua Decoction + sertraline group,which were better than that in the sertraline group.Conclusion Qinghua Decoction can improve the depressive like behavior of rats with syndrome of spleen-stomach dampness heat,and its mechanism may be related to increasing the content of ser-um 5-HT,restoring the morphological changes of hippocampal pathological tissue.
