Effect of Mongolian medicine Xiaozhong Jiuwei Powder on uric acid and inflammatory cytokines in rat with gouty arthritis
Objective It is to observe the effect of external application of Mongolian medicine Xiaozhong Jiuwei Powder on uric acid and inflammatory cytokines in rats with gouty arthritis(GA).Methods Forty male Wister rats were randomly divided into normal group,model group,Voltaren emulsion group,Yunnan Baiyao spray group,Mongolian medicine Xi-aozhong Jiuwei Powder group,with 8 rats in each group.The rats of all groups except for the normal group were given po-tassium oxonate and sodium urate to prepare models of GA.After successful modeling,the Voltaren emulsion group was given external application of Voltaren emulsion,the Yunnan Baiyao spray group was given external application of Yunnan Baiyao spray,and the Mongolian medicine Xiaozhong Jiuwei Powder group was given external application of Mongolian med-icine Xiaozhong Jiuwei Powder(mixed with egg white in the ratio of 1∶2 to form a paste),all once daily,continuously trea-ted for 14 days.The serum levels of uric acid(UA),tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2),inter-leukin-1(IL-1),interleukin-6(IL-6)and interleukin-8(IL-8)in the rats were determined,and the ankle joint tissue le-sions were observed by HE staining.Results After 14 days of treatment,the serum levels of UA,PGE2,IL-1,IL-6,IL-8,TNF-α in the rats of each treatment group were significantly lower than those of the model group(all P<0.05);the serum level of UA of the Voltaren emulsion group was significantly lower than those of the Yunnan Baiyao spray group and the Mongolian medicine Xiaozhong Jiuwei Powder group(all P<0.05),there was no statistically significant difference between the Yunnan Baiyao spray group and the Mongolian medicine Xiaozhong Jiuwei Powder group(P>0.05);the differences in the serum levels of PGE2 in the rats of each treatment group were not statistically significant(all P>0.05);the serum lev-els of IL-1,IL-8 and TNF-α in the rats of the Voltaren emulsion group and the serum levels of IL-6 and TNF-α of the Yun-nan Baiyao spray group were significantly lower than those of the Mongolian medicine Xiaozhong Jiuwei Powder group(all P<0.05);the serum level of IL-8 in the Mongolian medicine Xiaozhong Jiuwei Powder group was significantly lower than that in the Yunnan Baiyao spray group(P<0.05),and there was no statistically significant differences in the serum level of IL-1 compared with the Yunnan Baiyao spray group,and no statistically significant differences in the serum level of IL-6 compared with the Voltaren emulsion group(P>0.05).Compared with the model group,the overall structure of the ankle joint of rats in each treatment group was mildly abnormal,and the synovial inflammation was improved more or less,and the inflammatory cell infiltration and synovial hyperplasia were significantly alleviated,the improvement was the most obvi-ous in the Mongolian medicine Xiaozhong Jiuwei Powder group.Conclusion External application of Mongolian medicine Xi-aozhong Jiuwei Powder can effectively reduce the serum levels of UA and inflammatory cytokines,thus to alleviate inflam-mation injury in rats with gouty arthritis.
